摘要
目的:探讨COPD大鼠膈肌肌肉生长抑制素mRNA表达及其与膈肌凋亡萎缩的相关性。方法:8周龄雄性SD大鼠被随机分成正常对照组(n=8,假暴露)和COPD组(n=8,烟雾暴露),COPD组通过单纯熏香烟法复制大鼠COPD模型。检测支气管肺泡灌洗液细胞分类计数,RT-PCR检测膈肌肌肉生长抑制素mRNA表达、TUNEL法检测分析膈肌细胞凋亡指数。结果:与正常对照组相比,COPD组BALF中白细胞总数、中性粒细胞数、巨噬细胞数显著升高、大鼠体重和膈肌重量降低、膈肌细胞凋亡指数(AI)升高、膈肌中肌肉生长抑制素mRNA表达升高;COPD组膈肌中肌肉生长抑制素mRNA表达与膈肌重量均呈负相关,与膈肌细胞凋亡指数均呈正相关。结论:单纯熏香烟法成功建立大鼠COPD模型,COPD大鼠膈肌肌肉生长抑制素mRNA表达增加,可能与促进膈肌细胞凋亡及膈肌萎缩相关。
Objective To investigate the correlation between myostatin mRNA expression and apoptosis in diaphragm of COPD rats.Methods Male 8-wk-old rats were randomly divided into control group(n=8,rats undergoing sham exposure)and COPD group(n=8,rats undergoing cigarette smoke exposure).Cell type and count of bronchoalveolar lavage fluid were observed,myostatin mRNA expression was detected by RT-PCR,and the apoptotic index were measured and calculated by TUNEL.Results Compared with control group,the counts of white blood cells,neutrophils and macrophages in BALF increased in COPD group,the weight of body and diaphragm reduced in COPD group,the myostatin mRNA expression and apoptotic index of diaphragm rose in COPD group.Positive correlation was found between myostatin mRNA expression and apoptotic index of diaphragm,and negative correlation was found between myostatin mRNA expression and weight of diaphragm.Conclusion The COPD rats model was established by cigarette smoke exposure,myostatin mRNA expression of diaphragm in COPD rats significantly increased,that might be relevant to the apoptosis and atrophy of diaphragm.
