摘要
目的探讨红背叶根水提物对乙醇诱导的酒精性脂肪肝细胞的干预作用和可能机制。方法(1)采用不同浓度乙醇干预人肝L02细胞,检测细胞存活率以筛选适宜的建模浓度。(2)将L02细胞分为空白对照组和不同浓度红背叶根组,不同浓度红背叶根组经适宜浓度乙醇干预建模后,给予不同浓度红背叶根水提物干预,检测细胞存活率以筛选适宜的药物干预浓度。(3)将L02细胞分为空白对照组和0 mg/mL、0.78 mg/mL、1.56 mg/mL、3.125mg/mL红背叶根组,红背叶根组经适宜浓度乙醇干预建模后,给予相应浓度红背叶根水提物干预,检测并比较各组细胞的三酰甘油水平及陷窝蛋白1(Cav-1)mRNA表达水平。结果(1)使用80%的乙醇用于建模,将0.78 mg/mL、1.56 mg/mL、3.125 mg/mL作为红背叶根干预浓度。(2)各红背叶根组的三酰甘油水平及Cav-1 mRNA表达水平均高于空白对照组,且0 mg/mL、0.78 mg/mL、1.56 mg/mL、3.125 mg/mL红背叶根组三酰甘油水平及Cav-1 mRNA表达水平依次降低(均P<0.05)。结论红背叶根水提物可能通过降低三酰甘油水平及Cav-1 mRNA表达,发挥防治酒精性脂肪肝的作用。
Objective To investigate the intervention effect and potential mechanism of aqueous extracts from Radix Alchorneae Trewioidis on ethanol-induced alcoholic fatty liver cells.Methods(1)Human hepatic L02 cells were intervened using different concentrations of ethanol to measure cell viability so as to screen the optimum concentration for modeling.(2)L02 cells were divided into blank control group and different concentration Radix Alchorneae Trewioidis groups,different concentration Radix Alchorneae Trewioidis groups were intervened by different concentrations of aqueous extracts from Radix Alchorneae Trewioidis following the modeling treated with the optimum concentration of ethanol.The cell viability was measured to screen the optimum intervention concentration of the drug.(3)L02 cells were divided into blank control group,0 mg/mL,0.78 mg/mL,1.56 mg/mL and 3.125 mg/mL Radix Alchorneae Trewioidis groups,the Radix Alchorneae Trewioidis groups were treated with corresponding concentrations of aqueous extracts from Radix Alchorneae Trewioidis following the modeling treated with the optimum concentration of ethanol.TG level and caveolin(Cav)-1 mRNA expression level were detected and compared among groups.Results(1)Ethanol(80%)was applied to modeling,and 0.78 mg/mL,1.56 mg/mL,and 3.125 mg/mL represented the concentrations of Radix Alchorneae Trewioidis intervention.(2)The Radix Alchorneae Trewioidis groups exhibited higher TG level and Cav-1 mRNA expression level than the blank control group,moreover,TG level and Cav-1 mRNA expression level decreased in the sequence of the 0 mg/mL,0.78 mg/mL,1.56 mg/mL and 3.125 mg/mL Radix Alchorneae Trewioidis groups(all P<0.05).Conclusion Aqueous extracts from Radix Alchorneae Trewioidis play a role in the prevention and treatment of alcoholic fatty liver probably by reducing TC level and Cav-1 mRNA expression.
