摘要
该研究采用活泼酯法制备了人工抗原,通过动物免疫获得了针对2,4-二氯苯氧乙酸的特异性抗体。通过试验条件优化,建立了微孔侧流免疫层析方法,用于水果和蔬菜中2,4-二氯苯氧乙酸残留的快速检测。该方法的检测限:2,4-二氯苯氧乙酸为0.05μg/mL,4-氯苯氧乙酸为0.2μg/mL,2,4,5-三氯苯氧乙酸为0.5μg/mL。实际样品的检测中,其结果与HPLC的结果相一致率为100%。检测方法前处理简单、快速灵敏,且能够同时检测2,4-二氯苯氧乙酸及其结构类似物,适用于水果蔬菜中2,4-二氯苯氧乙酸残留的快速筛选。
The coating antigen and immunogen of the 2,4-dichlorophenoxyacetic acid(2,4-D)were prepared by the active ester method,respectively.The polyclonal antibody was obtained by immunizing rabbits,which could simultaneously recognize 2,4-dichlorophenoxyacetic acid.A microwell lateral flow immunochromatographic assay(mwLFIA)was developed for the fast detection of 2,4-D in agriculture products.The LOD of mwLFIA for 2,4-dichlorophenoxyacetic acid,4-Chlorophenoxyacetic acid and 2,4,5-trichlorophenoxyacetic acid were 0.05μg/mL,0.2μg/mL and 0.5μg/mL respectively.And it was accordance with HPLC results.The developed mwLFIA strip was suitable for the rapid detection of 2,4-dichlorophenoxyacetic acid and structural analogs in agriculture products.
