蒜氨酸抗细菌感染机制初探 被引量:4

Preliminary Discussion of Alliin to Treat Bacterial Infection and Its Mechanism

摘要 目的:以家兔脓肿模型,探讨蒜氨酸抗细菌感染机制。方法:将60只家兔脓肿模型,分为模型对照组、青霉素4×104 U/kg组、蒜氨酸0.5、1、5、10 mg/kg组,家兔静脉注射给予相应药物或生理盐水,连续7日。流式细胞术检测血液吞噬细胞吞噬率,酶联免疫吸附试验(ELISA)检测血清IL-2、IL-7含量,试管法检测家兔血清抗金葡菌抗体效价,流式细胞术检测脓汁标本中活菌率和死菌率。结果:经过一周治疗,模型对照组只有3只家兔存活,脓肿仍然存在,治疗组家兔均完全康复。与模型对照组比较,蒜氨酸各治疗组,脓汁标本死菌率显著增高(P<0.01);脓汁标本活菌率显著降低(P<0.01)。蒜氨酸各治疗组吞噬细胞吞噬率明显低于正常参考值,显著高于模型对照组(P<0.05),蒜氨酸治疗组血清IL-2、IL-7和抗金葡菌抗体均显著高于模型对照组和青霉素治疗组(P<0.01)。结论:蒜氨酸进入体内杀菌效果能与青霉素媲美,同时蒜氨酸提高吞噬细胞吞噬功能、提高机体免疫防御机能,具有双重抗感染效应。 Objective: Based on rabbit abscess model, to discuss the anti-infection mechanism of alliin. Methods: 60 abscess model rabbits were divided into the model group, 4×104U/kg penicillin group and 4 doses of alliin groups(0.5、1、5、10 mg/kg), all rabbits were intravenously administered with corresponding drugs or saline solution, for 7 days. The phagocytosis rate of phagocytes was detected by flow cytometry, the contents of IL-2 and IL-7 in serum were detected by enzyme-linked immunosorbent assay(ELISA). The titer of rabbit anti-staphylococcus aureus antibody was determined by test tube method, while the living bacteria rate and bacteria death rate in pus were detected by flow cytometry.Results: After a week of treatment, 3 rabbits with abscess survived in the model group, while all rabbits in other treatment groups survived and recovered. Compared with the model group, the bacteria death rates in all Alliin treatment groups were significantly increased(P<0.01), the living bacterium rates in all alliin treatment groups were significantly decreased(P<0.01). The phagocytosis rates in all alliin treatment groups were significantly lower than the normal reference value, but were significantly higher than that in the model group(P<0.05). Compared with penicillin group and the model group, the contents of serum IL-2, IL-7 and Rabbit anti-staphylococcus aureus antibody were significantly increased in all alliin treatment groups(P<0.01).Conclusion: Alliin has a comparable in vivo bactericidal effect with penicillin. At the same time, alliin may improve the phagocytic function, the body immune and the defense function, with dual anti-infection effect.
