摘要
目的观察急性胰腺炎大鼠半胱天冬酶-1(caspase-1)依赖性腺泡细胞焦亡的情况。方法选用Wistar大鼠12只,随机分为对照组和胰腺炎组各6只。胰腺炎组经腹腔注射精氨酸制作急性胰腺炎模型。造模成功后24 h,观察胰腺组织病理变化。采用蛋白免疫印迹法(Western blot)检测胰腺caspase-1、蛋白质gasdermin D(GSDMD)表达情况。采用酶联免疫分析(ELISA)方法检测造模成功后24 h血清白介素-1β(IL-1β)、白介素-18(IL-18)水平。并对胰腺caspase-1与血清IL-1β、IL-18水平进行Pearson相关性分析。结果病理检查显示,胰腺炎组大鼠胰腺组织以间质水肿、空泡形成及炎性浸润为主,对照组无明显病理改变;胰腺炎组与对照组大鼠的胰腺caspase-1[(0.59±0.04 vs 0.20±0.02)]、GSDMD[(0.58±0.08 vs 0.23±0.02)]表达水平比较,胰腺炎组明显高于对照组,差异均有统计学意义(P<0.05);血清ELISA检测显示:胰腺炎组与对照组大鼠的血清IL-1β[(115.14±13.12)pg/mL vs(24.34±4.52)pg/m L]、IL-18[(185.91±14.72)pg/m L vs(21.67±3.98)pg/m L]比较,胰腺炎组明显高于对照组,差异均有统计学意义(P<0.05);胰腺caspase-1与血清IL-1β、IL-18水平均呈正相关性(r=0.979、r=0.974,P<0.01)。结论在急性胰腺炎中存在显著的caspase-1依赖性腺泡细胞焦亡现象。
Objective To observe the pyroptosis of caspase-1-dependent acinar cells in rats with acute pancreatitis.Methods A total of 12 Wistar rats were randomly divided into two groups of 6 animals each:control group and pancreatitis group.Rats in the pancreatitis group were injected with L-arginine into the abdominal cavity to make acute pancreatitis models.24 hours after successful modeling,the histopathological changes of pancreas were observed.Western blot was used to detect the expression of caspase-1 and GSDMD in pancreas.ELISA was used to detect the serum levels of IL-1βand IL-18 at 24 h after the successful modeling.Pearson correlation analysis was performed between pancreatic caspase-1 and serum levels of IL-1βand IL-18.Results Pathological examination showed that slight pathological damage in pancreatitis group was mainly composed of interstitial edema,cavitation formation and inflammatory infiltration,while the control group had no obvious pathological changes.Western blot analysis showed that the expression levels of caspase-1 and GSDMD were significantly higher in the pancreatitis group than in the control group:caspase-1,(0.59±0.04)vs(0.20±0.02),P<0.05;GSDMD,(0.58±0.08)vs(0.23±0.02),P<0.05.ELISA showed that the levels of IL-1βand IL-18 were significantly higher in the pancreatitis group than in the control group:IL-1β,(115.14±13.12)pg/mL vs(24.34±4.52)pg/mL,P<0.05;IL-18,(185.91±14.72)pg/mL vs(21.67±3.98)pg/mL,P<0.05.Pancreatic caspase-1 was positively correlated with serum IL-1βand IL-18 levels(r=0.979,r=0.974,P<0.01).Conclusion There is significant pyroptosis of caspase-1-dependent acinar cell in acute pancreatitis.
