褐藻胶寡糖对白内障大鼠晶状体氧化损伤的保护作用及相关机制

摘要 目的探讨褐藻胶寡糖(AOS)对白内障大鼠晶状体氧化损伤的保护作用,并分析其可能机制。方法选择4周龄无特定病原体级SD雌性大鼠40只,取30只大鼠建立白内障模型,剩余10只大鼠给予常规饲料喂养。采用D-半乳糖诱导白内障大鼠模型。取建模成功的白内障模型大鼠24只,根据随机数字表法分为3组,分别为模型对照组、阳性对照组和AOS组,每组各8只。取同期正常饲料喂养的大鼠作为正常对照组,给予100 mg/kg 0.9%氯化钠溶液灌胃,模型对照组给予100 mg/kg 0.9%氯化钠溶液灌胃,阳性对照组给予30 mg/kg复明胶囊悬浊液灌胃,AOS组给予100 mg/kgAOS灌胃,每日1次,均连续治疗30 d后。在裂隙灯下观察大鼠晶状体组织浑浊程度并进行分级,然后计算完全浑浊率;分别检测大鼠血清和晶状体中氧化应激指标丙二醛(MDA)和一氧化氮(NO)含量以及总超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性;蛋白质印迹法检测大鼠晶状体中凋亡相关蛋白活化的含半胱氨酸的天冬氨酸蛋白水解酶-3(cleaved-Caspase-3)、Bcl-2和Bax的相对表达量。结果与正常组相比,模型对照组、阳性对照组和AOS组大鼠晶状体完全浑浊率、氧化指标MDA和NO含量以及凋亡指标cleaved-Caspase-3和Bax相对表达量均显著增加,而抗氧化指标SOD和GSH-Px活性和抗凋亡指标Bcl-2相对表达量均显著降低,差异均有统计学意义(P<0.05),与模型对照组相比,阳性对照组和AOS组大鼠晶状体完全浑浊率、氧化指标MDA和NO含量以及凋亡指标cleaved-Caspase-3和Bax相对表达量均显著降低,而抗氧化指标SOD和GSH-Px活性和抗凋亡指标Bcl-2相对表达量均显著增加,差异均有统计学意义(P<0.05),而与阳性对照组相比,AOS组以上指标差异均无统计学意义(P>0.05)。结论AOS干预后,白内障大晶状体浑浊程度显著改善,其机制可能与改善大鼠氧化应激反应水平,并提升大鼠抗氧化、抗凋亡能� Objective To investigate the protective effect of alginic oligosaccharides(AOS)on cataract lens oxidative damage and analyze its possible mechanism.Methods Forty specific pathogen free-grade SD female rats about 4 weeks old were selected,30 rats were selected to adopt D-galactose-induced cataract model,and the remaining 10 rats were selected as the normal control group and fed with regular diet.Twenty-four successfully modeled cataract model rats were divided into 3 groups according to the random number table method,namely the model control group,the positive control group and the AOS group,with 8 rats in each group.The normal control group was given 100 mg/kg normal saline by gavage,the model control group was given 100 mg/kg normal saline by gavage,the positive control group was given 30 mg/kg fuming capsule suspension by gavage,AOS group was given 100 mg/kg AOS by gavage,once a day,after 30 days of continuous treatment.The degree of turbidity of the rat lens tissue under the slit lamp were observed and graded,and then the contents of oxidative stress indicators of malondialdehyde(MDA)and nitric oxide(NO)in the serum and lens of rats,as well as the total superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities were detected;the relative expression of apoptosis-related proteins cleaved cysteinyl aspartate specific proteinase-3(cleaved-Caspase-3),Bcl-2 and Bax in rat lens was detected by Western blotting.Results Compared with the normal group,the model control group,positive control group and algin oligosaccharide group rat lens complete opacity rate,oxidation index MDA and NO content,and apoptosis index cleaved-Caspase-3 and Bax relative expression levels all increased significantly,while the anti-oxidation index SOD and GSH-Px activity and the relative expression of anti-apoptosis index Bcl-2 were significantly reduced,the differences were statistically significant(P<0.05);Compared with the model control group,the positive control group and the alginate oligosaccharide group rat lens complete opaci
