摘要
目的建立前臂同种异体移植的大鼠模型。方法在Brown Norway(BN)大鼠和Lewis大鼠间进行前臂的同种异体移植手术(n=8)。供者大鼠的腋动静脉与受者大鼠的颈外静脉、颈总动脉进行吻合,同时,尺神经、桡神经、正中神经进行吻合。对其中5只受者大鼠术后给予环孢素A进行免疫抑制治疗(观察组),3只受者大鼠未给予免疫抑制治疗(对照组)。术后评估移植前臂的存活情况、免疫排斥反应情况、感觉和运动的恢复情况。术后90 d进行移植物组织学观察。结果所有大鼠均能耐受此移植手术。在接受免疫抑制治疗的大鼠中,移植物长期存活,且受者移植前臂逐渐恢复运动和感觉功能,术后90 d时未见免疫排斥反应。在未接受免疫抑制治疗的大鼠中,移植物在术后12 d起逐渐发生坏死。结论前臂同种异体移植大鼠模型的制备是可行的。
Objective To establish a rat model of orthotopic forelimb allotransplantation.Methods Eight forelimbs were transplanted from Brown Norway rats to Lewis rats.Axillary vessels of transplant were used as the vascular pedicles,which were anastomosed to the external jugular vein and common carotid artery of the recipient rat.The ulnar,radial,and median nerves were also anastomosed.Five of the recipient rats received immunosuppressive therapy with cyclosporine A(observation group),while three received no immunosuppressive therapy(control group).The viability,signs of rejection and sensorimotor recovery of the transplanted forelimb were assessed after operation.Histological observation of the grafts were performed 90 days after operation.Results All rats survived after operation.In the rats treated with cyclosporine,the transplanted forelimbs achieved long-term survival with motion and sensation recovery.No immune rejection was observed 90 days after operation.In the rats without cyclosporine treatment,the transplanted forelimbs experienced tissue necrosis started from day 12 postoperatively.Conclusion The orthotopic forelimb allotransplantation in the rat model is feasible.
