Establishment and evaluation of a rat model of benign prostatic hyperplasia

摘要 目的建立非去势SD大鼠良性前列腺增生(BPH)模型,明确皮下注射丙酸睾酮(TP)具有诱导大鼠前列腺增生的作用,考察合适的诱导剂量,评估模型的维持时间及其对肾功能、体质量和各脏器指数的影响。方法将TP诱导组的大鼠前列腺指数和前列腺组织病理学切片与对照组进行比较。对比对照组和TP高、中、低3个剂量组血清双氢睾酮(DHT)、胰岛素样生长因子-1(IGF-1)、表皮生长因子(EGF)和BCL-2相关X蛋白(Bax)表达水平,并记录肾功能指标尿素氮(UREA)、肌酐(CREA)和血糖(GLU)的变化情况。以造模4周完成当天(实验第4周)和造模完成后4周(实验第8周)为2个时间点,对模型组与对照组大鼠前列腺指数及DHT表达水平进行比较,确定维持时间;并对各脏器指数及每周的体质量变化进行检测。结果与对照组相比,TP高剂量组组织病理形态和前列腺指数显示明显增生,DHT、IGF-1和EGF的表达水平显著升高,Bax表达水平显著降低。TP高剂量组大鼠CREA和GLU的表达水平与对照组比较差异有显著统计学意义。第4周TP高剂量组大鼠DHT表达水平、前列腺指数、肾脏指数和睾丸指数均显著高于对照组,胸腺和肝指数显著低于对照组;第8周时,仅前列腺和肾脏指数显著高于对照组,其他脏器及DHT表达水平均恢复正常。结论采用皮下注射丙酸睾酮50 mg·kg^(-1),2 d 1次,连续4周可成功诱导非去势SD大鼠BPH模型,该模型维持时间少于28 d,可引起大鼠体质量及多项脏器指数异常,使得肾指数升高并对肾功能产生不利影响。 Objective To establish a noncastrated SD rat benign prostatic hyperplasia(BPH)model and to determine the effect of subcutaneous injection of testosterone propionate(TP)on prostatic hyperplasia in rats,to investigate the appropriate induction dose,and to evaluate the maintenance time of the model and its effect on renal function,body weight and various organ indexes.Methods Prostatic index and prostate histopathological sections were compared between the TP induced groups and the control group.The serum dihydrotestosterone(DHT),insulin-like growth factor-1(IGF-1),epidermal growth factor(EGF),and BCL-2 associated X protein(Bax)expression levels were compared among the control group and TP high,middle and low dose groups.Renal function indexes of urea nitrogen(UREA),creatinine(CREA),and glucose(GLU)were recorded.2 time points were the day when the model was completed at the 4th week and after the model was maintained for 4 weeks(the 8th week),the prostate index and DHT levels of the model group and the control group were compared to determine the maintenance time,and the index of each organ with weekly changes in body mass were recorded.Results Compared with the control group,the histopathological morphology and prostate index of the TP high dose group showed obvious hyperplasia,and the expressions of DHT,IGF-1 and EGF were significantly increased,while the expressions of Bax were significantly decreased.The expression of CREA and GLU in the TP high dose group was significantly different from that in the control group.At the 4th week,DHT expression,prostatic index,renal index and testicular index in the TP high dose group were significantly higher than those in the control group,and thymus index and liver index were significantly lower than those in the control group.At the 8th week,only prostate index and kidney index were significantly higher than those of the control group,and the expression levels of DHT in other organs and other organs were restored to normal levels.Conclusion The BPH model of noncastrated SD r
