摘要
为探明不同产蛋阶段鹅等级前卵泡中FOXO3基因与凋亡和自噬相关基因的mRNA表达情况,采用荧光定量PCR方法检测开产前、产蛋初期和产蛋高峰期鹅等级前卵泡中FOXO3基因及Bcl-2、Bax、Caspase3、Fas、Fasl、LC3、Beclin1等凋亡和自噬相关基因mRNA的表达情况。结果显示:凋亡和自噬相关基因在鹅产蛋周期不同阶段等级前卵泡中均有表达;产蛋高峰期FOXO3、Bcl2、Bax、Caspase3、Fas和Fasl的mRNA表达量随卵泡直径变大呈增加趋势,LC3和Beclin1的表达量随卵泡直径变大呈减少趋势;产蛋初期各基因的表达变化不一致。由此可见,鹅等级前卵泡中FOXO3基因的表达随着卵泡的发育而增加,且其表达变化趋势与凋亡相关基因基本一致,但与自噬相关基因的变化趋势相反,推测FOXO3基因可能与鹅卵泡颗粒细胞的凋亡和自噬有一定关联。
To explore FOXO3 gene and apoptosis and autophagy related genes expression in pre-hierarchical follicles of goose at different laying period, the mRNA expression of FOXO3 gene and apoptotic and autophagy-related genes including Bcl-2, Bax, Caspase-3, Fas, Fasl, LC3 and Beclin1 in pre-hierarchical follicles of geese during pre-laying,early-laying and peak-laying periods were detected by qRT-PCR. The results showed that these genes were expressed in pre-hierarchical follicles during different stages of the egg-laying cycle. During the peak-laying period, the mRNA expression of FOXO3, Bcl2, Bax, Caspase3, Fas and Fasl increased with the increase of follicular diameter, while the mRNA expression of LC3 and Beclin1 decreased with the increase of follicular diameter. During early-laying period,the expression patterns of these genes were not consistent. This study demonstrated that the expression of FOXO3 gene increased with the development of follicles, and the change trend of FOXO3 gene expression was consistent with that of apoptosis related genes, but opposite to that of autophagy related genes. It suggested that FOXO3 gene might be associated with apoptosis and autophagy of goose follicular granulosa cells.
