摘要
目的:建立绞股蓝药材高效液相色谱-紫外-蒸发光散射器(HPLC-UV-ELSD)串联指纹图谱方法,并同时测定槲皮素、芦丁、绞股蓝皂苷A和绞股蓝皂苷XLIX的含量,对其质量优劣进行全面综合评价。方法:采用HPLC-UV-ELSD串联法和相似度评价软件,对15批不同产地绞股蓝药材进行指纹图谱研究,并对绞股蓝中槲皮素、芦丁、绞股蓝皂苷A和绞股蓝皂苷XLIX 4种指标性成分中的含量进行测定与分析。结果:建立了绞股蓝药材HPLCUV-ELSD串联指纹图谱法,标定了11个共有峰,指认了其中4个色谱峰;不同产地绞股蓝药材中槲皮素、芦丁、绞股蓝皂苷A和绞股蓝皂苷XLIX 4种成分的含量高低不一,质量存在差异。结论:建立了绞股蓝药材HPLC-UVELSD指纹图谱法,并结合相似度评价和4种成分的定量分析结果,较全面地反映该药材的质量,该法可用于不同产地绞股蓝药材质量的有效评价。
Objective:To establish a HPLC-UV-ELSD fingerprint method for simultaneous determination of the content of quercetin,rutin,gypenoside A and gypenoside XLIX in Jiaogulan(Gynostemma pentaphyllum).Methods:HPLC-UV-ELSD tandem method and similarity evaluation software were used to study the fingerprints of 15 batches of Jiaogulan(Gynostemma pentaphyllum)from different origins,and the contents of quercetin,rutin,gypenoside A and gypenoside XLIX in Jiaogulan(Gynostemma pentaphyllum)were determined and analyzed.Results:The HPLC-UV-ELSD tandem fingerprint method of Jiaogulan(Gynostemma pentaphyllum)was established,11 common peaks were calibrated,and 4 chromatographic peaks were identified.The content of quercetin,rutin,gypenoside A and gypenoside XLIX in different origins of Jiaogulan(Gynostemma pentaphyllum)were different,and the quality was different.Conclusion:The HPLC-UV-ELSD fingerprinting method of Jiaogulan(Gynostemma pentaphyllum)was established,and the quality of Jiaogulan(Gynostemma pentaphyllum)was comprehensively reflected by the similarity evaluation and the quantitative analysis of four components.This method can be used to effectively evaluate the quality of Jiaogulan(Gynostemma pentaphyllum)from different origins.
