摘要 目的探讨双功能小分子多肽P7(序列LPLTPLP)联合替莫唑胺(TMZ)对人胶质母细胞瘤细胞系U87MG的影响。方法以TMZ与P7单用或联用处理U87MG细胞,采用CCK-8法检测细胞活性,流式细胞术检测U87MG细胞凋亡水平,蛋白质免疫印迹法检测U87MG细胞凋亡相关蛋白水平。以溶剂作对照。结果TMZ和P7均能抑制U87MG细胞的生长,当P7与TMZ浓度比为1∶1时,协同作用最明显;与对照组比较,联合给药组的U87MG细胞凋亡率显著升高(P<0.01),cleaved caspase-9和cleaved caspase-3表达水平显著升高(P<0.05)。结论P7联合TMZ能协同抑制U87MG细胞活力,并促进其凋亡,其机制可能与显著上调促凋亡蛋白cleaved caspase-9和cleaved caspase-3水平有关。 Objective To investigate the effect of bifunctional polypeptide P7(LPLTPLP)combined with temozolomide(TMZ)on the human glioblastoma cell line U87 MG in vitro.Methods U87 MG cells were treated with TMZ and P7 alone or in combination,and the cell viability was detected by CCK-8 assay.The apoptosis level of U87 MG cells was detected by flow cytometry.The expression of apoptosis-related proteins was detected by western blot.The solvent was used as the control group.Results Both TMZ and P7 could inhibit the growth of U87 MG cells.When the concentration ratio of P7 to TMZ was 1∶1,the synergistic effect was the most obvious.Compared with those in the control group,the apoptosis rate in the combined group was significantly increased(P<0.01),and the expression levels of cleaved caspase-9 and cleaved caspase-3 in the combined group were significantly increased(P<0.05).Conclusion P7 combined with TMZ can synergistically inhibit the survival of U87 MG cells and promote their apoptosis,which may be related to the up-regulation of pro-apoptotic protein(cleaved caspase-9 and cleaved caspase-3)levels.
