筇竹Na+/H+逆向转运蛋白基因克隆与表达分析 被引量:4

Cloning and Expression Analysis on QtNHX1 Gene from Qiongzhuea tumidinoda

摘要 为明确液泡型Na+/H+逆向转运蛋白在竹类植物耐盐中发挥的重要作用。本研究从筇竹中克隆出液泡膜Na+/H+逆向转运蛋白基因,命名为QtNHX1(基因登录号:MT078987),并分析该基因表达特征。结果表明,QtNHX1基因全长为2075 bp,包含1632 bp的开放阅读框(编码544个氨基酸),其相对分子量为59.43 kD,理论等电点(pI)为8.83。QtNHX1具有10个跨膜区域,且与水稻同源性最高,达到90.59%。另外,QtNHX1基因在在筇竹不同组织器官(竹笋,根,茎,叶)均有表达,说明QtNHX1为组成型表达。筇竹根中QtNHX1基因的表达量最高,在竹笋表达量最低。对筇竹进行不同时间(6,12,24 h)的NaCl胁迫处理,发现QtNHX1基因的表达量随着处理时间的延长而上升,表明QtNHX1基因在调控筇竹耐盐性中发挥着重要作用。 In order to study the role of vacuolar Na+/H+antiporter in bamboo plants under salt tolerance.The sfuty cloned a vacuolar Na+/H+antiporter gene,named QtNHX1,from Qiongzhuea tumidinoda and its expression pattern in different tissues and under salt treatment were analyzed.The results showed that the full length of QtNHX1 gene was 2075 bp,and the open reading frame(ORF)of QtNHX1 was 1632 bp,encoding 544 amino acids.The relative molecular weight of QtNHX1 was 59.43 kD,with the theoretical isoelectric point 8.83.QtNHX1 protein had 10 transmembrane regions,and had the highest homology with rice(90.5%).In addition,QtNHX1 gene was expressed in bamboo shoots,roots,stems and leaves without NaCl treatment,indicating that QtNHX1 was non-induced expression.The expression level of QtNHX1 was the highest in the roots and the lowest in shoots.Under NaCl stress,the expression of QtNHX1 gene increased with the increase of treatment time(6,12,24 h).The results suggested that QtNHX1 gene was closely related to salt tolerance of Qiongzhuea tumidinoda.
