摘要
目的探讨miR-34a对肾癌细胞增殖、侵袭的调控机制及其与患者预后的相关性.方法选择2019年4月至2020年3月手术切除获得的肾癌组织细胞、癌旁组织细胞标本30例,经荧光定量PCR法检测肾癌组织细胞内miR-34a表达,经MTT法检测miR-34a抑制肾癌miR-34a细胞增殖、侵袭机制,并采用Kaplan-Meier生存模型分析miR-34a与患者预后的相关性.结果肾癌组织细胞内miR-34a相对表达量分别是(0.84±0.13)、(1.78±0.22),肾癌组织细胞内miR-34a相对表达量低于癌旁组织,差异有统计学意义(t=20.148,P<0.01);细胞转染培养24 h、48 h后,两组细胞吸光度值均明显升高,但miR-34a高表达组低于miR-34a低表达组(P<0.05);细胞转染培养24 h后,miR-34a高表达组肾癌侵袭细胞数为(87.14±9.23),明显低于miR-34a低表达组的肾癌侵袭细胞数(142.89±11.44)(P<0.05);miR-34a高表达组患者无复发生存率明显高于miR-34a低表达组(P<0.05).结论miR-34a可对肾癌组织细胞增殖、侵袭能力进行抑制,加快肿瘤细胞凋亡,提高患者预后生存质量,可作为肾癌临床治疗、预后评估的新靶点.
Objective To investigate the expression of miR-34a in kidney cancer cells and its regulation mechanism on the proliferation and invasion of renal cancer cells,and to explore its correlation with the prognosis of patients.Methods 30 cases of specimens of kidney cancer tissue cells and adjacent cancer tssue cells obtained from our hospital From April 2019 to March 2020 were selected as the research materials.Fluorescence quantitative PCR method was used to detect the expression of miR-34a in renal cell carcinoma cells.The mechanism of miR-34a inhibition of proliferation and invasion of renal cancer miR-34a cells was detected by MTT method,and the Kaplan-Meier survival model was used to analyze the correlation between miR-34a and patient prognosis.Results The relative expression levels of miR-34a in renal cancer tissue cells were(0.84+0.13)and(1.78+0.22).The relative expression levels of miR-34a in renal cancer tssue cells were lower than those in adjacent tissues,the difference was statistically significant(t=20.148,P<0.01).After the cells were transfected and cultured for 24 h and 48 h,the absorbance values of the two groups of cells increased significantly,but the miR-34a high expression group was lower than the miR--34a low expression group(P<0.05).After 24 hours of cell transfection and culture,the number of invasive cells of kidney cancer in the miR-34a high expression group was(87.14+9.23),which was significantly lower than the number of invasive cells of kidney cancer in the miR-34a low expression group(142.89+11.44)(P<0.05).The relapse-free survival rate of miR.-34a high expression group was significantly higher than that of miR-34a low expression group(P<0.05).Conclusion miR:-34a can inhibit the proliferation and invasion of kidney cancer tsses,accelerate tumor cell apoptosis,improve the prognostic quality of life of patients,and can be used as a new target for clinical treatment and prognosis evaluation of kidney cancer.
