摘要
目的研究nexilin F-肌动蛋白结合蛋白(NEXN)基因表达与结直肠癌患者预后及免疫细胞浸润的相关性。方法利用Oncomine数据库分析NEXN基因在肿瘤和正常组织中的表达差异;通过Prognoscan数据库分析NEXN基因表达与结直肠癌患者预后的关系;通过TIMER及GEPIA数据库分析NEXN基因高表达与免疫浸润的相关性。结果 Oncomine数据库分析结果表明NEXN在结直肠癌中高表达(fold change=3.057,P<0.000 1);Prognoscan数据库GSE17536和GSE14333的芯片分析结果表明,NEXN高表达与结直肠癌患者的不良预后呈正相关(P<0.05);TIMER数据库分析显示NEXN与CD4+T细胞、巨噬细胞、树突状细胞及中性粒细胞的浸润具有中度或强的相关性(COR值>0.4,P<0.05);进一步分析不同细胞亚群标志基因发现NEXN高表达与Treg细胞、M2巨噬细胞标志基因表达呈中度或强的相关性,与T细胞耗竭相关基因TIM3和TIGIT呈中度相关性(COR值>0.4,P <0.05);利用GEPIA数据库分析也证实上述结果。结论 NEXN高表达与结直肠癌患者不良预后呈正相关,而M2型巨噬细胞、Treg细胞的浸润可能是重要的原因。
This study was designed to evaluate the correlation of nexilin F-actin binding protein(NEXN) gene expression with the prognosis and immune cell infiltration of patients with colorectal cancer. NEXN gene expression was analyzed via the Oncomine database,and the relationship between NEXN gene expression and prognosis of colorectal cancer patients was analyzed by Prognoscan database;the correlation between NEXN gene and immune infiltrating cell marker gene was analyzed by Timer and GEPIA database. Oncomine database analysis showed that NEXN were highly expressed in colorectal cancer tissues. The cohorts(GSE17536 and GSE14333) of colorectal cancer patients showed that high NEXN expression was positively correlated with the poor prognosis(P<0.05). And NEXN expression was moderately or strongly correlated with the infiltration of CD4+T cells, macrophages, dendritic cells and neutrophils(COR>0.4, P<0.05). Further analysis of the marker genes of different cell subsets showed that the expression of NEXN was moderately or strongly correlated with the expression of Treg cell marker gene and M2macrophage marker gene(COR>0.4, P<0.05). Takentogether, the high expression of NEXN in colorectalcancer patients is positively correlated with the poorprognosis, and the infiltration of M2 macrophages andTreg cells play an important role in this process.
