摘要
目的基于网络药理学研究枳实薤白桂枝汤和人参汤治疗皆主胸痹的科学内涵。方法利用TCMID、TCMSP数据库,筛选枳实薤白桂枝汤和人参汤的有效成分,并应用Swiss Target Prediction和Drugbank数据库检索作用靶点,通过Cytoscape得到两复方的共同活性成分以及共同靶点。利用HGMD、TTD、OMIM、DisGeNET、DrugBank和HPO 6个疾病数据库筛选心绞痛的相关靶点,与两复方的靶点取得交集,得到两复方治疗心绞痛的共有靶点和特有靶点。将上述靶点输入String数据库进行蛋白质—蛋白质相互作用网络预测,利用R3.6.1软件进行GO生物过程以及KEGG通路分析。结果筛选出枳实薤白桂枝汤和人参汤的药物活性成分数分别为43个和111个,药物靶点数分别为200个和212个,共有药物活性成分和靶点数分别为4个和183个。通过作用靶点分析,两复方治疗心绞痛的作用靶点基本来自三种共有的活性成分,作用机理可能存在高度相似性。蛋白质相互作用网络分析,两复方治疗心绞痛可能与白细胞介素(interlenkin,IL)-6、血管内皮生长因子(vascular endothelial growth factor,VEGF)、趋化因子配体2(chemokine C-C motif ligand 2,CCL2)、IL-10、IL-1B、SERPINE1等靶点相关。GO以及KEGG分析结果显示:两复方治疗心绞痛与低氧诱导因子-1(hypoxia-inducible factor 1,HIF-1)信号通路、IL-17信号通路、细胞因子及其受体的相互作用、核因子κB(nuclear factor kappa-B,NF-κB)信号通路密切相关。结论枳实薤白桂枝汤和人参汤中共有靶点及靶点所在的生物过程和通路,可以共同治疗心绞痛。而枳实薤白桂枝汤特有靶点(金属蛋白酶组织抑制因子1)和人参汤特有靶点(单胺氧化酶A)作用的不同,可能是枳实薤白桂枝汤与人参汤皆主胸痹的生物学实质。
Objective To study the scientific connotation of Zhishi Xiebai Guizhi decoction and Renshen decoction both treating obstruction Qi in the chest based on the scientific connotation.Methods TCMID and TCMSP database were used to screen the active components of Zhishi Xiebai Guizhi decoction and Renshen decoction,and Swiss Target Prediction and Drugbank database were used to search the targets.The common active components and targets of Zhishi Xiebai Guizhi decoction and Renshen decoction were obtained by Cytoscape.6 disease databases,including HGMD,TTD,OMIM,DisGeNET,DrugBank and HPO,were used to screen the relevant targets of angina pectoris,and the intersects with the targets of the two compounds were obtained,so as to obtain the common and specific targets of the two compounds for treating angina pectoris.The above targets were input into the String database for protein-protein interaction network prediction,and R3.6.1 software was used to analyze the biological process of GO and KEGG pathway.Results A total of 43 and 111 active components in Zhishi Xiebai Guizhi decoction and Renshen decoction were identified respectively,and 200 and 212 targets were identified,respectively.A total of 4 common active components and 183 targets were identified,respectively.By analyzing the targets,the targets of the two compounds in the treatment of angina pectoris are basically from three common active components,and the mechanism of action may be highly similar.Protein interaction network analysis indicated that the two compounds play a role in treating angina pecpecis may be related to IL-6,VEGFA,CCl2,IL-10,IL-1B,SERPINE1 and other targets.The results of GO and KEGG analysis showed that the two compounds play a role in treating angina pectoris was closely related to HIF-1,IL-17 and NF-κB signaling pathways,the interaction of cytokines and their receptors.Conclusion Zhishi Xiebai Guizhi decoction and Renshen decoction can treat angina pectoris together,by their common targets and the biological processes and pathways of targets
