基于网络药理学分析复方鬼针草颗粒治疗高血压的作用机制 被引量:1

摘要 目的:通过网络药理学方法研究复方鬼针草颗粒的活性成分、作用靶点及治疗高血压可能的作用机制。方法:采用TCMSP数据库及Swiss Target Prediction数据库获取复方鬼针草颗粒的活性成分及作用靶点,利用Uniprot数据库对靶点进行标准化,通过GeneCard、OMIM数据库获得高血压的疾病靶点。将药物作用靶点与疾病靶点的交集作为药物-疾病的共同靶点。应用STRING数据库获得蛋白-蛋白相互作用关系,通过Cytoscape3.7.2软件进行可视化筛选核心靶点,并绘制出“中药-活性成分-靶点-疾病”网络,利用DAVID数据库对这些共同作用靶点进行分析和KEGG分析。结果:筛选出复方鬼针草颗粒94个活性成分,146个与高血压相关的共同作用靶点。核心靶点涉及血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)、RAC-α丝氨酸/苏氨酸蛋白激酶(RAC-alpha serine/threonine-protein kinase,AKT1)、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子(tumor necrosis factor,TNF)等。GO分析共富集出854个条目,其中血管收缩正调控、细胞增殖正调控、凋亡过程负调控等生物过程与高血压密切相关,KEGG分析共映射出122条通路,其中TNF信号通路、缺氧诱导因子-1(hypoxia-inducible factor-1,HIF-1)信号通路、cGMP-PKG、PI3K-Akt、Toll样受体信号通路等是复方鬼针草颗粒治疗高血压相关度较高的通路。结论:复方鬼针草颗粒可能通过所含的槲皮素、山柰酚、木犀草素等活性成分与VEGFA、AKT1、IL-6、TNF等核心靶点相互作用,影响血管收缩、细胞增殖、凋亡等生物过程和调控TNF、HIF-1、cGMP-PKG、PI3K-Akt、Toll样受体等信号通路而发挥降压作用。 Objective:To study the active components,targets,and potential mechanism of compound Guizhencao(Bidens pilosa L.)Granules in hypertension treatment based on network pharmacology method.Methods:The active components and target of compound Guizhencao granules were obtained by using the TCMSP database and Swiss Target Prediction database,the targets were standardized by the Uniprot database,and the disease targets of hypertension were obtained via GeneCard and OMIM database.Then the intersection of drug action targets and disease targets were taken as the common drug-disease targets.The protein-protein interaction relationship was obtained by using the STRING database.Cytoscape3.7.2 software was used to perform visualization screening for core targets,and the"Traditional Chinese medicine-active components-targets-diseases"network was plotted.And GO analysis and KEGG analysis was performed on those common targets through the DAVID database.Results:94 active components and 146 targets related to hypertension were selected from compound Guizhencao granules.The core targets involved VEGFA,AKT1,IL-6,and TNF,etc.854 entries resulted from GO analysis of enrichment,of which the positive regulation of vasoconstriction,positive regulation of cell proliferation,and negative regulation of apoptosis are closely related to hypertension.122 pathways were reflected from KEGG analysis,of which the TNF,HIF-1,cGMP-PKG,PI3K-Akt,Toll-like receptors signaling pathways are highly relevant with pathways for compound Guizhencao granules treatment in hypertension.Conclusion:Compound Guizhencao Granules may interact with core targets such as VEGFA,AKT1,IL-6,and TNF through active ingredients such as quercetin,kaempferol,luteolin,etc.,and affect vasoconstriction,cell proliferation,apoptosis,and regulate TNF,HIF-1,cGMP-PKG,PI3K-Akt,Toll-like receptors,and other signaling pathways.And playing the role in lowering blood pressure through such biological processes.
