摘要
目的:研究补肾化瘀生新方在大鼠体内的入血成分,为其药效物质基础及作用机理研究奠定基础。方法:采用液相色谱-串联质谱仪,应用Thermo Accucore C18色谱柱(100 mm×2.1 mm,2.6μm),以甲醇(A)和0.1%甲酸水(B)为流动相,流速0.2 mL·min^(-1),梯度洗脱(0.00~5.00 min 95%→55%B,5.00~14.00 min 55%→37%B,14.00~26.00 min 37%→10%B,26.00~34.00 min 10%→0%B,34.00~35.00 min 0%B),柱温30℃,进样量5μL,检测波长254 nm,电喷雾离子源(ESI),DMSS2模式,正、负离子分别扫描,扫描范围m/z120~1200。对大鼠灌胃补肾化瘀生新方后的含药血清、空白血清及对照品溶液进行检测分析,结合文献相关信息,鉴别化合物结构,确定入血成分。结果:共鉴定出梓醇、地黄苦苷、甲基异茜草素、大黄素甲醚、黄芩苷、阿魏酸6个原型入血成分,这些成分主要来源于熟地黄、巴戟天、当归、川芎。结论:这些入血成分可能是补肾化瘀生新方在体内直接作用的物质,为阐明其药效物质基础和作用机制提供实验依据。
Objective:To explore the absorbed components of Bushen Huayu Shengxin Decoction,and building the foundation for its further pharmacodynamic material basis and mechanism studies.Methods:Liquid chromatography-tandem mass spectrometer and Thermo Accucore C18 column(100 mm×2.1 mm,2.6μm)was used,methanol(A)and 0.1%formic acid water(B)were used as mobile phases,with a flow rate of 0.2 mL·min^(-1),gradient elution(0.00-5.00min 95%→55%B,5.00-14.00 min 55%→37%B,14.00-26.00 min 37%→10%B,26.00-34.00 min 10%→0%B,34.00-35.00 min 0%B),column temperature 30℃,injection volume 5μL,detection wavelength 254 nm,electrospray ion source(ESI)at DMSS2 mode,positive and negative ions were scanned respectively,and the scanning range was m/z 120-1200.The drug-containing serum of Bushen Huayu Shengxin Decoction,the blank serum,and the reference solution was detected and analyzed.Then combined with the relevant literature,the structure of the compounds was identified,and the blood components were determined.Results:Six components of catalpol,Rehmapicrogenin,rubiadin,physcion,baicalin,and ferulic acid were identified in the drug-containing serum,all of which mainly came from Dihuang(Rehmannia glutinosa),Bajitian(Radix Morindae Officinalis),Danggui(Angelica Sinensis)and Chuangxiong(Ligusticum chuanxiong Hort).Conclusion:The absorbed components of Bushen Huayu Shengxin Decoction may act directly in the human body,which helps to clarify its pharmacodynamic material basis and mechanism.
