摘要
目的探讨半夏泻心汤治疗胃肠动力障碍性疾病的可能作用机制。方法30只SD大鼠随机分为半夏泻心汤组和去离子水组,每组15只,分别给予半夏泻心汤及去离子水12.15g/(kg·d)灌胃7天,制备半夏泻心汤药物血清及去离子水血清。小鼠胃Cajal间质细胞(ICC)分为空白组,去离子水血清组及5%、10%、15%半夏泻心汤血清组。空白组加入10%胎牛血清,其余各组加入相应药物血清,各组给药量均为2ml。检测ICC细胞凋亡情况,选择抑制ICC细胞凋亡的半夏泻心汤血清最优浓度进行后续试验。设空白组、去离子水血清组、半夏泻心汤血清组。空白组、去离子水血清组给药浓度同前,半夏泻心汤血清组给予筛选结果的半夏泻心汤血清浓度检测ICC细胞半胱氨酸天冬氨酸蛋白酶3(Caspase-3)、B淋巴细胞瘤2(Bcl-2)、Bcl-2相关x蛋白质(Bax)、胞内磷脂酰肌醇激酶(PI3K)蛋白表达,一氧化氮(NO)、一氧化氮合成酶(eNOS)含量。结果与空白组和去离子水血清组比较,10%半夏泻心汤血清组ICC细胞凋亡率均降低(P<0.05),且10%半夏泻心汤血清组ICC细胞凋亡率低于5%半夏泻心汤血清组和15%半夏泻心汤血清组(P<0.05),选择10%半夏泻心汤血清用于后续试验。与空白组和去离子水血清组比较,半夏泻心汤血清组ICC细胞Bcl-2蛋白表达显著升高,Caspase-3、Bax、PI3K蛋白表达,分泌NO、eNOS显著下降(P<0.05)。结论半夏泻心汤可能通过调控ICC细胞凋亡相关蛋白Caspase-3、Bax、PI3K、Bcl-2及抑制NO、eNOS分泌,抑制ICC细胞凋亡,从而发挥治疗胃肠动力障碍性疾病的作用。
Objective To explore the possible mechanism of Banxia Xiexin Decoction(BXD,半夏泻心汤) in the treatment of disorders of gastrointestinal motility(DGIM).Methods Thirty SD rats were randomly divided into BXD group and deionized water group with 15 rats in each group.Correspondingly,BXD and deionized water at a dose of 12.15 g/(kg·d)were given by gavage for seven days,so as to prepare the BXD-contained and deionized water-contained serum.Gastric interstitial cells of Cajal(ICC)of rats were divided into blank group,deionized water group,and 5%,10%and 15%BXD group.The blank group was given 10%fetal calf serum,while the other groups were given either deionized water-contained or BXD-contained serum correspondingly;the dose was 2 mL in all the groups.The apoptosis of ICC was assessed,and the result was used to figure out the optimum concentration of BXD for conducting a further test,which include the blank group,the deionized water group and BXD group.In this further test,the drug concentration was same as mentioned above except for the BXD group which used the obtained optimum concentration.The protein expression of cysteine-requiring aspartate protease 3(Caspase 3),B lymphocyte tumor 2(Bcl-2),Bcl-2-associated X protein(Bax),and intracellular phosphatidylinositol 3-kinase(PI3 K)was detected,and the nitric oxide(NO)and nitric oxide synthase(eNOS)content were assessed.Results The apoptosis rate of ICC in 10%BXD group was significantly lower than that of blank group and deionized water group(P<0.05),as well as that of 5%and 15%BXD group(P<0.05);therefore,10%BXD was selected to be used in the further test.Compared to blank group and deionized water group,the BXD group had higher protein expression of Bcl-2 of ICC,as well as lower protein expression of Caspase-3,BaX and PI3 Kand,and secretion of NO and eNOS(P<0.05).Conclusion BXD may play role in DGIM through inhibiting the apoptosis of ICC,and the mechanism may be related to its regulation of apoptosis-related proteins including Caspase-3,Bax,PI3 K and Bcl-2,and inhi
