摘要
目的探讨胶质母细胞瘤中基质金属蛋白酶组织抑制物1(TIMP1)、基质金属蛋白酶组织抑制物2(TIMP2)的表达及与微血管密度(MVD)的关系。方法收集42例接受手术治疗的胶质母细胞瘤患者的胶质母细胞瘤组织和周围正常组织。采用蛋白质印迹法检测两种组织中TIMP1、TIMP2的表达情况,采用免疫组化两步法检测两种组织的MVD。比较胶质母细胞瘤组织和周围正常组织中TIMP1、TIMP2的相对表达量及MVD值,比较不同组织学分级胶质母细胞瘤组织中TIMP1、TIMP2的相对表达量及MVD值,采用Pearson相关分析法分析胶质母细胞瘤组织中TIMP1、TIMP2表达与MVD的相关性。结果胶质母细胞瘤组织中TIMP1、TIMP2的相对表达量均明显低于周围正常组织,MVD值明显高于周围正常组织,差异均有统计学意义(P﹤0.01)。Ⅳ级胶质母细胞瘤组织中TIMP1、TIMP2的相对表达量均明显低于Ⅲ级胶质母细胞瘤组织,MVD值明显高于Ⅲ级胶质母细胞瘤组织,差异均有统计学意义(P﹤0.01)。胶质母细胞瘤组织中TIMP1、TIMP2的表达均与MVD呈负相关(r=-0.796、-0.815,P﹤0.01)。结论胶质母细胞瘤组织中TIMP1、TIMP2的表达水平明显低于周围正常组织,MVD值明显高于周围正常组织,且恶性程度越高,TIMP1和TIMP2的表达水平越低,MVD值越高,TIMP1、TIMP2的表达均与MVD值呈负相关。
Objective To investigate the expression of tissue inhibitor of metalloproteinase 1(TIMP1)and tissue inhibitor of metalloproteinase 2(TIMP2)in glioblastoma and their relationship with microvessel density(MVD).Method The glioblastoma tissues and surrounding normal tissues of 42 patients with glioblastoma undergoing surgical treatment were collected.The expression of TIMP1,TIMP2 in all tissues were detected by Western blotting.The MVD was measured by the two-step immunohistochemistry assay.The relative expression levels of TIMP1 and TIMP2 and MVD value in glioblastoma tissues and surrounding normal tissues were compared.The relative expression levels of TIMP1 and TIMP2 and MVD value in glioblastoma tissues of different histological grades were also compared.The correlation between TIMP1 and TIMP2 expression and MVD in glioblastoma tissues was analyzed by Pearson correlation analysis.Result The relative expression levels of TIMP1 and TIMP2 in glioblastoma tissues were significantly lower than those in the surrounding normal tissues,and the MVD was significantly higher than that in the surrounding normal tissues,and the differences were statistically significant(P<0.01).The relative expression levels of TIMP1 and TIMP2 in grade IV glioblastoma tissues were significantly lower than those in gradeⅢglioblastoma tissues,and the MVD value was significantly higher than that in gradeⅢglioblastoma tissues,the differences were statistically significant(P<0.01).The expressions of TIMP1 and TIMP2 in glioblastoma tissues were negatively correlated with the MVD(r=-0.796,-0.815,P<0.01).Conclusion The expression levels of TIMP1 and TIMP2 in glioblastoma tissues were significantly lower than those in the surrounding normal tissues,and the MVD value was significantly higher than that in the surrounding normal tissues.The expression level of TIMP1 and TIMP2 are lower,and the MVD value is higher with a more poor degree of malignancy.The expression of TIMP1 and TIMP2 is negatively correlated with the MVD value.
