摘要
从喜树中分离筛选出可以产喜树碱的内生真菌,并对其进行分子生物学鉴定。通过组织块内生菌分离纯化方法,自喜树中的叶、茎及果实中共分离得到45株内生真菌,对其进行了PDA培养基及喜树果煎汁PDA培养基发酵测试,筛选到两株可以在喜树果煎汁PDA培养基中产喜树碱及10-羟基喜树碱的内生真菌,产量最高分别为1.03 mg/g干菌丝和0.22 mg/g干菌丝。并对两株菌进行了分子生物学鉴定,结果确认两株内生真菌分别隶属于链格孢属及葡萄座腔菌属。
The paper separates the endophytic fungi which can produce camptothecin from camptotheca acuminate decne,and identifies it by methods of molecular biology.The total of 45 endophytic fungi was isolated from the leaves,stems and fruits of camptotheca acuminate decne with tissue culture method.Two endophytic fungi were screened by fermentation test with the potato glucose medium and the potato glucose medium added the juice of camptotheca acuminate decne fruit.The camptothecin and the 10-hydroxy camptothecin were found in the mycelium of two endophytic fungi.The yield of camptothecin was 1.03 mg/g dried mycelium and the yield of 10-hydroxy camptothecin was 0.22 mg/g dry mycelium tested with HPLC.We identified the two endophytic fungi by molecular biology methods and the results showed that they belonged to alternaria and botryosphaeria respectively.
