摘要
目的基于癌症基因组图谱(TCGA)数据库分析着丝粒蛋白A(CENPA)基因在肝细胞肝癌(HCC)中的表达情况以及临床意义。方法从TCGA数据库中通过筛选得到240例HCC患者和50例癌旁组织样本完整的临床病理及生存资料,评价CENPA基因在肝癌组织和癌旁组织中的表达情况。并根据CENPA基因中位表达水平将其分为低表达组(119例)和高表达组(121例),分析CENPA基因表达水平与HCC患者临床病理特征的关系以及CENPA基因表达水平、临床病理特征与HCC预后的相关性。结果与癌旁组织相比,CENPA基因在HCC癌组织中表达水平较高(P<0.05)。HCC癌组织中CENPA基因表达水平与HCC患者的性别、肿瘤病理学分级、血清AFP水平均相关(P<0.05)。多因素Cox回归分析显示,癌症家族史、TNM分期Ⅲ~Ⅳ期以及CENPA基因高表达是影响HCC患者预后的独立危险因素(P<0.05)。结论CENPA基因高表达与晚期肿瘤进展和不良预后显著相关,可以作为预测HCC患者临床结果的新型潜在生物标志物。
Objective To analyze the expression of centromere protein A(CENPA)gene and its clinical significance in hepatocellular carcinoma(HCC)based on the Cancer Genome Atlas(TCGA)database.Methods The complete clinicopathological and survival data of 240 patients with HCC and 50 paracancerous tissue samples were obtained by screening from TCGA database.The expression levels of CENPA gene in HCC tissues and paracancerous tissues were evaluated.They were divided into low expression group(119 cases)and high expression group(121 cases)according to the median expression level of CENPA gene.The relationship between the expression level of CENPA gene and clinicopathological features of patients with HCC were analyzed,and the correlations between the expression level of CENPA gene,clinicopathological features and prognosis of HCC were analyzed.Results The expression level of CENPA gene was higher in HCC tissues compared with the paracancerous tissues(P<0.05).The expression level of CENPA gene in HCC tissues was correlated with the gender,tumor pathological grade and serum alpha-fetoprotein(AFP)level of patients with HCC(P<0.05).Multivariate Cox regression analysis showed that family history of cancer,TNM stageⅢtoⅣand high expression level of CENPA gene were independent risk factors for the prognosis of patients with HCC(P<0.05).Conclusion The high expression level of CENPA gene is significantly associated with advanced tumor progression and poor prognosis.CENPA gene may function as a novel potential biomarker for predicting the clinical outcomes of patients with HCC.
