摘要 目的:研究骨形态发生蛋白-2(BMP-2)对放射性损伤后骨髓间充质干细胞(BMSCs)向成纤维分化的抑制作用及分子机制。方法:建立BMSCs细胞放射损伤模型,选择最佳BMP-2干预剂量;体外培养BMSCs,随机分为对照组、放射组、BMP-2组、BMP-2+头蛋白组(Noggin)。利用Western blot、免疫荧光技术和RT-PCR等方法检测不同组别α-平滑肌肌动蛋白(α-SMA)、Ⅰ型胶原(Col-Ⅰ)、Ⅲ型胶原(Col-Ⅲ)及转化生长因子β1(TGF-β1)、Smad2/3、p-Smad2/3等蛋白及分子的表达变化情况。结果:放射组BMSCs中α-SMA、Col-Ⅰ、Col-Ⅲ、TGF-β1及p-Smad2/3的表达较对照组明显增高(P<0.05),使用BMP-2干预后其表达量较放射组明显下降(P<0.05),Noggin组表达量与放射组无统计学差异。结论:BMP-2可抑制放射损伤后BMSCs向成纤维细胞分化,其机制可能与TGF-β1/Smad途径密切相关。 Objective:To study the inhibitory effects of BMP-2 on the differentiation of bone marrow mesenchymal stem cells(BMSCs)into myofibroblasts after radiation injury and to explore the related molecular mechanism.Methods:Radiation injury model of BMSCs was established.BMSCs were randomly divided into control group,radiation group(radiation alone),BMP-2 group(adding BMP-2 after radiation)and BMP-2+Noggin group.The expression ofα-smooth muscle actin(α-SMA),type I collagen(Col-Ⅰ),type III collagen(Col-Ⅲ),transforming growth factor-β1(TGF-β1),Smad2/3,p-Smad2/3 were detected in all groups by Western blot,immunofluorescence technique and RT-PCR respectively.Results:The expression ofα-SMA,Col-I,Col-III,TGF-β1 and p-Smad 2/3 in radiation group were significantly higher than that in control group(P<0.05).After the intervention of BMP-2,the expression ofα-SMA,Col-I,Col-III,TGF-β1 and p-Smad2/3 were decreased(P<0.05).However,there was no statistical difference between BMP-2+Noggin group and radiation group.Conclusion:BMP-2 can inhibit the differentiation of BMSCs into myofibroblasts after radiation injury,and its mechanism may be related to TGF-β1/Smads pathway.
