酸枣仁汤治疗失眠的功效网络与机制研究 被引量:7

摘要 目的:运用网络药理学方法探究酸枣仁汤治疗失眠的潜在机制。方法:应用中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)数据库筛选酸枣仁汤的组成药味成分并应用TCMSP、PubChem和Swiss Target Prediction数据库预测成分靶点。应用数据库Therapeutic Target Database(TTD)、Drugbank和Comparative Toxicogenomics Database(CTD)查找失眠靶点。靶点映射后借助Cytoscape软件构建成分-靶点(Compounds-Targets,C-T)网络和蛋白互作(Protein-Protein Interaction,PPI)网络,并拓扑关键靶点进行富集分析。结果:检索得到酸枣仁汤成分140个,成分作用靶点137个,其中治疗失眠的靶点77个,拓扑GABRA1、GABRA4、GABRB3、GABRA3、GABRB2等关键靶点30个。基因本体(Gene Ontology,GO)和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)富集分析主要涉及神经递质受体活性参与突触后膜电位的调节、对铵离子的反应、调节血管口径、G蛋白偶联胺受体活性、镉离子细胞反应、环化酶活性的正调节等生物过程,以及神经活性配体-受体相互作用、尼古丁成瘾、钙信号通路、肌萎缩侧索硬化症、Th1和Th2细胞分化、5-羟色胺能突触等信号通路。结论:酸枣仁汤中槲皮素、7-甲氧基-2-甲基异黄酮、山奈酚、豆甾醇、酸枣仁碱K等多种成分可能通过调控神经递质受体活性参与突触后膜电位的调节、G蛋白偶联胺受体活性等多生物过程,干预神经活性配体-受体相互作用、钙信号通路等多通路,发挥治疗失眠的作用。 Objective:To investigate the potential mechanisms of Suanzaoren decoction(酸枣仁汤)in the treatment of insomnia by using network pharmacology methods.Methods:The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)was used to screen the compounds of Suanzaoren decoction and at the same time,TCMSP,PubChem and Swiss Target Prediction were used to predict the targets.The Therapeutic Target Database(TTD),Drugbank and the Comparative Toxicogenomics Database(CTD)were used to find targets of insomnia.After targets mapping,Compounds-Targets(C-T)network and Protein-Protein Interaction(PPI)network were constructed by using Cytoscape,and enrichment analysis was performed on the key targets identified by the topology.Result:A total of 140 compounds of Suanzaoren decoction were screened,and there were 137 corresponding targets,which were contained in 77 targets associated with insomnia.30 core targets such as GABRA1,GABRA4,GABRB3,GABRA3 and GABRB2 were topologically identified.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis mainly involved the involvement of neurotransmitter receptor activity in the regulation of postsynaptic membrane potential,reaction to ammonium ions,regulation of vascular diameter,G-protein-coupled amine receptor activity,cadmium ion cell reaction,positive regulation of cyclase activity and other biological processes and neuroactive ligand-receptor interactions,nicotine addiction,calcium signaling pathways,amyotrophic lateral sclerosis,Th1 and Th2 cell differentiation,serotonin synapses,and other signaling pathways.Conclusion:The variety of compounds such as quercetin,7-Methoxy-2-methyl isoflavone,kaempferol,Stigmasterol and(S)-Coclaurine in Suanzaoren decoction may play a role in the treatment of insomnia by regulating the activity of neurotransmitter receptors,it participates in the regulation of postsynaptic membrane potential,G-protein-coupled amine receptor activity and other biological processes,and interferes with the neur
