摘要
旨在探究丹参提取物是否可以提高特定抗生素对金黄色葡萄球菌(包括耐甲氧西林金黄色葡萄球菌)的抗菌活性。通过棋盘法最小抑菌浓度检测、生长曲线和杀菌曲线等试验确定丹参提取物及其有效成分与不同抗生素的体外协同作用,进一步通过LDH试验和细胞保护试验验证丹参提取物的细胞毒性及其与庆大霉素在细胞水平上的协同效果,最后通过高效液相色谱法对丹参提取物的有效成分进行检测和分析。结果显示,丹参提取物显著提高氨基糖苷类抗生素、β-内酰胺类抗生素和多黏菌素等抗生素的体外抗金黄色葡萄球菌活性,丹参提取物同样可显著提高庆大霉素对不同细胞由金黄色葡萄球菌介导损伤的保护作用。丹参提取物主要成分包括隐丹参酮、丹参酮Ⅰ、丹参酮ⅡA和丹酚酸B等,其中隐丹参酮与庆大霉素协同效果最为显著,其可能是丹参提取物发挥作用的有效成分之一,但丹参提取物中其他成分是否发挥更佳的协同作用有待进一步研究。因此,丹参提取物作为庆大霉素增效剂具有潜在的开发价值。
The aim of this study was to investigate whether the extract of Salvia miltiorrhiza could enhance the antibacterial activity of different antibiotics against S.aureus.The synergistic effects of the extract of Salvia miltiorrhiza and its ingredients with different antibiotics in vitro were determined by the checkerboard minimum inhibitory concentration(MIC)test,growth curve test and time-killing assay.The synergistic effects of the extract of Salvia miltiorrhiza and gentamicin at the cellular level were further verified by the LDH test and rhodopsin test.The results showed that the extract of Salvia miltiorrhiza significantly increased the antibacterial activity of aminoglycoside antibiotics,β-lactam antibiotics and polymyxins against S.aureus in vitro,and the extract of Salvia miltiorrhiza also increased the protective effect of gentamicin against S.aureusmediated injury at the cellular level.The synergistic effect of the active ingredients of the extract of Salvia miltiorrhiza,such as cryptotanshinone with gentamicin was found to be significant,but the synergistic effect of other components in the extract of Salvia miltiorrhiza needs further study.
