大鼠急性心肌梗死区域miRNA-29和miRNA-21的动态变化与心功能的关系 被引量:4

The relationship between the dynamic changes of miRNA-29 and miRNA-21 in acute myocardial infarction area and cardiac function in rats

摘要 目的探讨大鼠急性心肌梗死后早期梗死区域miRNA-29和miRNA-21的动态变化与心功能的关系。方法 SD雄性大鼠25只,结扎冠状动脉前降支,建立大鼠心肌梗死模型,分为术后7、14、21、28 d组和假手术组(对照组),每组各5只。彩色超声心动图检测大鼠左室射血分数(LVEF)、左室短轴缩短率(LVFS)和左室舒张末期内径(LVIDd)。石蜡切片,苏木精-伊红(HE)和Masson染色。Western blotting检测大鼠心肌梗死后梗死区的胶原纤维I(I)和弹性蛋白(E)的变化。实时荧光定量聚合酶链法检测大鼠心肌梗死区域miRNA-29和miRNA-21的水平。结果大鼠心肌梗死后,心功能下降,2周时LVEF和LVFS下降至最低,后缓慢升高(P<0.05);胶原I逐渐升高(P<0.01),弹性蛋白E逐渐降低(P<0.05)。而且较对照组,心肌梗死区miRNA-29于第1周上升明显,第2周迅速下降(P<0.01);miRNA-21第2周明显上升,后逐渐下降(P<0.01)。结论 miRNA-29和miRNA-21表达升高可能在急性心肌梗死早期的心功能降低中起重要作用。 Objective To explore the relationship between the dynamic changes of miRNA-29 and miRNA-21 in the infarct area in the early infarction area and the cardiac function after acute myocardial infarction in rats.Methods Twenty-five SD male rats were selected.The rat model of myocardial infarction was established by ligating the anterior descending coronary artery.All rats were divided into 7 d group,14 d group,21 d group,28 d group and the sham operation group(control group)with 5rats in each group.Left ventricular ejection fraction(LVEF),left ventricular short axis shortening rate(LVFS),and left ventricular end diastolic diameter(LVIDd)were measured by color echocardiography.Paraffin sections of heart tissue were stained with HE and Masson.The changes of collagen fiber I(Collagen fiber I,I)and elastin(Elastin,E)in infarcted area after myocardial infarction were detected by Western blotting.Real-time fluorescent quantitative polymerase chain reaction was used to detected the level of mi RNA-29 and mi RNA-21 in myocardial infarction area.Results Cardiac function declines after myocardial infarction in rats.LVEF and LVFS went down to the lowest level in the second week and then slowly improved(P<0.05).Collagen I gradually increased(P<0.01),and elastin E gradually decreased(P<0.05).Compared with the control group,the level of mi RNA-29 in the myocardial infarction area increased significantly in one week,then decreased rapidly in the second week(P<0.01).While miRNA-21 rose visiblely in the second week and then declined gradually till the following two weeks(P<0.01).Conclusion The increased expression of miRNA-29 and miRNA-21 may play an important role in the reduction of heart function in the early stage of acute myocardial infarction.
