摘要
内质网应激是内质网错误折叠或未折叠蛋白的异常聚集引起的内质网功能紊乱。近几年的研究发现内质网应激在肝脏疾病中发挥重要作用。肝衰竭是临床上高病死率疾病,病情进展迅速,发病机制复杂。目前关于内质网应激是否参与肝衰竭的发生发展暂无系统性认识。本文主要对内质网应激的概念、信号通道及其与肝细胞代谢、炎症、氧化应激的研究作一概述,并归纳了内质网应激在肝衰竭动物模型和临床中的最新研究结果,以便更好地了解内质网应激在肝衰竭中的作用。
Endoplasmic reticulum stress is the pathologic process of accumulation of misfolded or unfolded proteins leading to functional disturbance of endoplasmic reticulum.Recent studies indicate that endoplasmic reticulum plays an important role in liver diseases.Liver failure is a clinical disease with high mortality rate,rapid disease progression,and complex pathogenesis.At present,there is still no systematic understanding whether endoplasmic reticulum involve in the development and progression of liver failure.This article mainly reviewed the concept,signaling pathway of endoplasmic reticulum and its relationship with hepatocyte metabolism,inflammation and oxidative stress,and the latest research results of endoplasmic reticulum in animal models and clinical trials of liver failure,in order to better understand the role of endoplasmic reticulum stress in liver failure.
