LMP基因多态性与脊髓灰质炎疫苗诱导抗体应答的相关性

摘要 目的探讨低相对分子量蛋白酶体(LMP)基因单核苷酸多态性位点(SNPs)与脊髓灰质炎疫苗序贯免疫诱导的脊髓灰质炎病毒抗体应答的相关性分析。方法选取357名2~3月龄的婴幼儿接种2剂灭活脊髓灰质炎疫苗(IPV,由Ⅰ、Ⅱ、Ⅲ三类脊灰病毒灭活制成)和1剂二价口服脊髓灰质炎疫苗(bOPV),于疫苗免疫前和基础免疫后第28天时检测婴幼儿血清中3种型别的脊髓灰质炎病毒中和抗体水平;采用TaqMan探针基因分型法对LMP 2基因SNPs位点rs17587(G>A)和LMP 7基因rs2071543(G>T)进行基因分型,分析这2个SNPs位点与脊髓灰质炎病毒抗体应答的相关性。结果Ⅰ、Ⅱ、Ⅲ型脊髓灰质炎病毒中和抗体几何平均滴度(GMT)水平在携带rs17587和rs2071543位点不同等位基因的个体间比较,差异无统计学意义(P>0.05);rs17587和rs2071543不同基因型携带者的Ⅰ、Ⅱ、Ⅲ型脊髓灰质炎病毒中和抗体GMT水平比较,差异无统计学意义(P>0.05);rs17587-AA基因型携带者的脊髓灰质炎病毒Ⅱ型中和抗体GMT水平高于AG和GG基因型携带者(P<0.05),但经Bonferroni校正后,rs17587-AA基因型携带者的脊髓灰质炎病毒Ⅱ型中和抗体GMT水平与GG基因型携带者比较,差异无统计学意义(P>0.05)。结论LMP基因的SNPs位点rs2071543和rs17587与脊髓灰质炎疫苗免疫后抗体应答无相关性,不是影响脊髓灰质炎疫苗抗体应答的主要遗传因素。 Objective To evaluate the association of single nucleotide polymorphism(SNPs) in low molecular weight polypeptide(LMP) gene with poliovirus vaccine-induced antibody response.Methods A total of 357 infants aged 2-3 months were selected to receive two doses of inactivated poliomyelitis vaccine(IPV,made by the inactivation of three serotypes of poliovirus Ⅰ,Ⅱ,and Ⅲ)and one dose of bivalent oral poliomyelitis vaccine(bOPV). The levels of neutralizing antibodies against three types of poliovirus in serum of infants were detected before immunization and 28 days after basic immunization. The genotyping of LMP2 gene rs17587(G > A) and LMP7 gene rs2071543(G >T) was performed by TaqMan SNP genotyping assay. The correlation between SNPs and poliovirus vaccine-induced antibody response was analyzed. Results There was no significant difference in GMT levels of polio type Ⅰ,Ⅱ,and Ⅲ antibodies among individuals with different alleles of rs17587 and rs2071543(P > 0. 05). There was no statistically significant difference in GMT of polio type Ⅰ,Ⅱ,and Ⅲ antibodies in carriers with different genotypes of rs17587 and rs2071543(P > 0. 05). Polio type Ⅱ GMT of the rs17587-AA genotype carriers were higher than those of the AG and GG genotype carriers(P < 0. 05). However,after Bonferroni correction,there was no significant difference in the GMT level of neutralizing antibody against poliovirus type Ⅱ in carriers of rs17587-AA genotype compared with carriers of GG genotype(P > 0. 05). Conclusion SNPs rs2071543 and rs17587 of the LMP gene are not significantly correlated with polio vaccine-induced antibody response and not the main genetic factors affecting the antibody response of polio.
