摘要
目的:探讨艾拉莫德对干燥综合征小鼠颌下腺炎症及NF-κB信号通路的影响,阐明艾拉莫德对干燥综合征小鼠的可能作用机制。方法:将36只NOD小鼠根据随机数字法分为模型组(M组)、艾拉莫德1组(I1组)、艾拉莫德2组(I2组),12只/组,12只C57BL/6小鼠作为对照组(C组)。I1组小鼠每天给予羧甲基纤维素+10 mg/kg艾拉莫德灌胃治疗,I2组小鼠每天给予羧甲基纤维素+30 mg/kg艾拉莫德灌胃治疗,C组和M组小鼠每天给予等量羧甲基纤维素灌胃,共8周。8周末,测定各组小鼠饮水量、唾液分泌量、颌下腺指数和脾脏指数,HE染色测定颌下腺病理形态学变化。ELISA测定颌下腺组织IL-6、TNF-α、IFN-γ水平。Western blot测定下颌腺组织NF-κB/p50、NF-κB/p65、IκBα蛋白水平。结果:与C组相比,M组小鼠每日饮水量升高(P<0.05),唾液分泌量降低(P<0.05),颌下腺指数降低(P<0.05),脾脏指数升高(P<0.05),颌下腺组织IL-6、TNF-α、IFN-γ水平升高(P<0.05),下颌腺组织NF-κB/p50、NF-κB/p65蛋白水平升高(P<0.05),IκBα蛋白水平降低(P<0.05);与M组相比,I组小鼠每日饮水量降低(P<0.05),唾液分泌量升高(P<0.05),颌下腺指数升高(P<0.05),脾脏指数降低(P<0.05),颌下腺组织IL-6、TNF-α、IFN-γ水平降低(P<0.05),下颌腺组织NF-κB/p50、NF-κB/p65蛋白水平降低(P<0.05),IκBα蛋白水平升高(P<0.05)。与I1组相比,I2组各指标变化更显著(P<0.05)。结论:艾拉莫德可抑制干燥综合征小鼠下颌腺的炎症反应和NF-κB信号通路激活,对干燥综合征小鼠下颌腺发挥保护作用。
Objective:To investigate the effect of iguratimod on inflammation and NF-κB signaling pathway of the submandibular gland in mice with Sjogren’s syndrome in order to elucidate the possible mechanism of action of iguratimod on mice with Sjogren’ssyndrome.Methods:36 NOD mice were divided into model group(M group),iguratimod 1 group(I1 group),and ifuratimod 2 group(I2 group)accorded to random number method,with 12 rats in each group.12 C57BL/6 mice served as control group(C Group).I1 group mice were given carboxymethylcellulose+10 mg/kg iguratimod daily for treatment.I2 group mice were given carboxymethylcellulose+30 mg/kg iguratimod daily for treatment.C group and M group mice were given equal amount of carboxymethylcellulose per day for8 weeks.At the end of the 8 th week,the water consumption,salivary secretion,submandibular gland index and spleen index of eachgroup of mice were determined.Pathological changes of submandibular gland were determined by HE staining.Levels of IL-6,TNF-αand IFN-γin submandibular gland were determined by ELISA.Western blot was used to detect levels of NF-κB/p50,NF-κB/p65 andIκBαin submandibular gland.Results:Compared with C group,the daily drinking water was increased(P<0.05),saliva secretion wasdecreased(P<0.05),submandibular gland index was decreased(P<0.05),spleen index was increased(P<0.05),levels of IL-6,TNF-αand IFN-γin submandibular gland were increased(P<0.05),levels of NF-κB/p50 and NF-κB/p65 in submandibular gland were increased(P<0.05),and level of IκBαprotein was decreased(P<0.05)in M group mice.Compared with M group,the daily drinking water was decreased(P<0.05),saliva secretion was increased(P<0.05),submandibular gland index was increased(P<0.05),spleen index was decreased(P<0.05),levels of IL-6,TNF-αand IFN-γin submandibular gland were decreased(P<0.05),levels of NF-κB/p50 and NF-κB/p65 in submandibular gland were decreased(P<0.05),and level of IκBαprotein was increased(P<0.05)in I groupmice.Compared with I1 group,changes of each indexs in the I2 gr
