摘要
目的探讨紧密连接蛋白(Claudin-5)、核转录因子(NF-κB)、细胞周期抑制蛋白(P21)在老化型C57BL/6J小鼠耳蜗外侧壁血管纹内的表达及意义。方法将C57BL/6J小鼠分为青年组(3月龄)和老年组(24月龄),每组各20只,检测两组小鼠听性脑干反应(ABR)的反应阈,分别行组织学切片观察耳蜗外侧壁血管纹形态结构,免疫组化检测Claudin-5、NF-κB、P21在血管纹表达,反转录聚合酶链反应(RT-PCR)检测Claudin-5、NF-κB、P21在耳蜗的表达水平。结果与青年组小鼠相比,老年组C57BL/6J小鼠ABR阈值升高,差异有统计学意义(均为P<0.05);老年组C57BL/6J小鼠血管纹内细胞上Claudin-5表达减少,NF-κB、P21表达增多,差异有统计学意义(均为P<0.01);RT-PCR检测显示老年组耳蜗内Claudin-5含量明显减少,NF-κB、P21含量明显增多,差异均有统计学意义(均为P<0.01)。结论Claudin-5、NF-κB、P21在老年组C57BL/6J小鼠耳蜗外侧壁血管纹内均有表达,Claudin-5表达降低可能与血管纹通透性相关,NF-κB和P21表达增多可能与内外淋巴循环紊乱和细胞凋亡有关。
Objective To study the expression of Claudin-5,NF-κB and P21 in the stria vascularis of the outer cochlea of aging C57BL/6J mice.Methods C57BL/6J mice were divided into 3-month-old group and 24-month-old group with 20 mice in each group.The hearing level of the two groups was measured by auditory brainstem response(ABR).The morphological structure of the stria vascularis of the lateral wall of the cochlea was observed by histological section,the expression of Claudin-5,NF-κB and P21 in the stria vascularis was detected by immunohistochemistry,and the expression of Claudin-5,NF-κB and P21 in the cochlea was detected by RT-PCR.Results①Compared with young mice,the ABR threshold of old C57BL/6J mice increased significantly(P<0.05).The expression of Claudin-5 in stria vascularis of C57BL/6J mice in the old group were decreased,and the expression of NF-κB and P21 was increased(P<0.01).②Compared with young mice,RT-PCR results showed the expression of Claudin-5 in the cochlea was decreased and the expression of NF-κB and P21 was increased(P<0.01).Conclusion Claudin-5,NF-κB and P21 are all expressed in the stria vascularis of the lateral wall of the cochlea of C57BL/6J mice in the old group.The low expression of Claudin-5 may be related to the permeability of stria vascularis.The increased expression of NF-κB and P21 may be related to the disorder of internal and external lymphatic circulation and apoptosis.
