类叶牡丹提取物经NF-κB信号通路对类风湿关节炎滑膜细胞凋亡与炎症因子的作用机制 被引量:7

摘要 目的:研究类叶牡丹提取物(CRME)经NF-κB信号通路对类风湿关节炎(RA)滑膜细胞凋亡与炎症因子的作用机制。方法:纳入2019年1-12月于本院接受关节清理术治疗的RA患者25例进行研究。采集所有患者的滑膜组织4份,分成模型组、低剂量组、中剂量组及高剂量组,各25份。另取同期于本院接受诊治的25例非RA患者滑膜组织作为对照组。其中模型组及对照组均不予以任何处理,低、中、高剂量组分别予以50、100、500 μg/ml的CRME进行处理。采用四甲基偶氮唑蓝比色法(MTT)检测各组滑膜细胞凋亡情况,以酶联免疫吸附法(ELISA)检测各组IL-6、IL-4、VEGF及Bax、FasL、Bcl-2表达水平,采用Western boltting法检测p50、p65、IκBα水平。结果:模型组、低剂量组、中剂量组、高剂量组细胞抑制率均呈逐渐升高趋势,差异有统计学意义(P<0.05)。模型组细胞IL-4、IL-6、VEGF水平均显著高于对照组,低剂量组、中剂量组、高剂量组IL-4水平均显著高于模型组,IL-6、VEGF水平均显著低于模型组,差异均有统计学意义(P<0.05)。模型组细胞Bax、FasL、Bcl-2水平均显著高于对照组,低剂量组、中剂量组、高剂量组Bax、FasL水平均显著低于模型组(P<0.05),Bcl-2水平高于模型组(P<0.05)。模型组细胞p50、p65、IκBα表达水平显著高于对照组,且低剂量组、中剂量组、高剂量组p50、p65、IκBα表达水平显著低于模型组(P<0.05)。结论:CRME主要是通过调节NF-κB信号通路,从而抑制炎症因子、抗凋亡因子的表达,同时促进促凋亡因子的表达,进一步发挥治疗RA的作用。 Objective:To study the mechanism of caulophyllum robustum maxim extract (CRME) by NF-κB signaling pathway on apoptosis and inflammatory factors in synovium cells of rheumatoid arthritis (RA) through NF-κB signaling pathway.Method:A total of 25 RA patients who received arthroplasty in our hospital from January to December 2019 were included in the study.Four synovial tissues of all patients were collected and divided into the model group,the low dose group,the medium dose group and the high dose group,with 25 samples each.In addition,synovial tissues of 25 non-RA patients receiving treatment in our hospital during the same period were taken as the control group.The model group and the control group were not treated,while the low,medium and high dose groups were treated with CRME of 50,100 and 500 μg/ml,respectively.The apoptosis of synovial cells in each group was detected by tetramethylazolium blue colorimetry (MTT).The expression levels of IL-6,IL-4,VEGF,Bax,FasL and Bcl-2 were detected by enzyme-linked immunosorbent assay (ELISA).The levels of p50,p65 and IκBα were detected by Western boltting assay.Result:The cell inhibition rates of the model group,the low-dose group,the medium-dose group and the high-dose group all showed a trend of gradual increase,the differences were statistically significant (P<0.05).The levels of IL-4,IL-6 and VEGF in the model group were significantly higher than those in the control group;the levels of IL-4 in the low-dose group,the medium-dose group and the high-dose group were significantly higher than those in the model group,while the levels of IL-6 and VEGF were significantly lower than those in the model group (P<0.05).The levels of Bax,FasL and Bcl-2 in the model group were significantly higher than those in the control group,while the levels of Bax and FasL in the low-dose,medium-dose and high-dose groups were significantly lower than those in the model group (P<0.05),the Bcl-2 level was higher than that in the model group (P<0.05).The expression levels of p50,p65 and Iκ
