摘要
目的:通过研究疏肝调神针法对偏头痛大鼠行为学、外周血中5-羟色胺(5-hydroxytryptamine,5-HT)、降钙素基因相关肽(calcitonin gene-related peptide,CGRP)、P物质(substance P,SP)及三叉神经脊束核中腺苷A1受体(adenosine A1 receptor,ADORA1)基因和蛋白表达的影响,探讨本法治疗偏头痛的作用机制。方法:将32只雄性Wistar大鼠按随机数字表法分为对照组、模型组、普通针刺组、疏肝调神组,采用颈后皮下注射硝酸甘油制备偏头痛模型,每天固定大鼠30 min,疏肝调神组选取风池(双)、百会、太冲(双)、内关(双),普通针刺组取百会、风池(双)进行针刺,余组只抓取固定,连续8 d。采用ELISA法检测血中5-HT、CGRP、SP含量,荧光定量PCR法、免疫组化法检测三叉神经脊束核中ADORA1的mRNA、蛋白表达情况。结果:与对照组比较,模型组血清5-HT含量明显降低,CGRP、SP含量明显升高;与模型组比较,疏肝调神组和普通针刺组5-HT含量明显升高,CGRP、SP含量明显降低;与普通针刺组比较,疏肝调神组大鼠血中5-HT含量明显升高,CGRP、SP的含量明显降低;与对照组比较,模型组大鼠三叉神经脊束核中ADORA1的mRNA、免疫组化阳性表达明显降低;与模型组比较,疏肝调神组、普通针刺组大鼠三叉神经脊束核中ADORA1的mRNA、免疫组化阳性表达均明显升高;与普通针刺组比较,疏肝调神组大鼠三叉神经脊束核中ADORA1的mRNA、免疫组化阳性表达明显升高。结论:疏肝调神针法可调节大鼠三叉神经脊束核中ADORA1及血中5-HT、CGRP、SP含量并发挥镇痛作用,且相较于普通针刺作用更明显。
Objective:To observe the ecffects of soothing liver and regulating shen acupuncture therapy on expression of 5-hydroxytryptamine(5-HT),Calcitonin Gene-Related Peptide(CGRP),substance P(SP)and Adenosine A1 Receptor(ADORA1)in migraine rats,so as to explore the possible mechanism of the therapy in treatment of migraine.Methods:32 male Wistar rats were randomly divided into control group,model group,soothing liver and regulating shen group and ordinary acupuncture group(n=8 in each group).Migraine model was established by hypodermic injection of glyceryl trinitrate.Rats in each group were immobilized for 30 min for 8 days.At the time of immobilizing,Baihui(DU 20),Fengchi(GB 20),Neiguan(PC 6)and Taichong(LR 3)were selected for acupuncture in soothing liver and regulating shen group,Baihui(DU 20)and Fengchi(GB 20)were selected for acupuncture in ordinary acupuncture group.Expression of 5-HT,CGRP,SP and ADORA1 were detected by ELISA,fluorescent quantitative PCR and immunohistochemistry respectively.Results:Compared with the control group,the expression of 5-HT and ADORA1 were significantly decreased and the expression of CGRP and SP were significantly increased in the model group.Compared with the model group,the expression of 5-HT and ADORA1 were significantly increased,CGRP and SP were significantly decreased in soothing liver and regulating shen group and the ordinary acupuncture group.Compared with ordinary acupuncture group,the expression of 5-HT and ADORA1 were significantly increased while CGRP and SP were significantly decreased in soothing liver and regulating shen group.Conclusion:Acupuncture therapy of soothing liver and regulating shen can regulate the expression of 5-HT,CGRP,SP and ADORA1 in migraine rats and relieve migraine effectively.And this therapy is more effective than ordinary acupuncture in relieving migraine.
