基于血清代谢组学探究茵陈蒿汤对非酒精性脂肪肝大鼠的影响 被引量:7

Study on the effect of Yinchenhao decoction on non-alcoholic Fatty liver Model Rats based on Serum Metabonomics

摘要 目的采用血清代谢组学的方法探究茵陈蒿汤防治非酒精性脂肪肝(NAFLD)的作用机制,并通过寻找具有生物学意义的内源性潜在生物标志物,以此探寻其相关代谢通路。方法采用高脂饮食诱导大鼠建立NAFLD模型,造模成功后,给予3.6g/kg茵陈蒿汤进行治疗,连续2周。应用UPLC-Q-TOF-MS技术对空白组、模型组与茵陈蒿汤组大鼠血清代谢组学进行分析,采用主成分分析法探寻潜在的关键性生物标志物,联合人类代谢组学数据库(HMDB)和京都基因与基因组百科全书数据库(KEGG)找出对应的相关性代谢通路。结果茵陈蒿汤可改善NAFLD模型大鼠代谢轨迹的偏离,同时寻找出与NAFLD模型及给予茵陈蒿汤干预后具有共同差异的生物标志物11个,涉及10条相关的代谢通路,应用相关代谢通路分析发现,影响最为广泛的四条代谢通路可能是甘油磷脂代谢、亚油酸代谢、α-亚麻酸代谢及烟碱和烟碱胺代谢。结论茵陈蒿汤对NAFLD模型大鼠血清代谢产物具有调节作用,为进一步对茵陈蒿汤治疗相关疾病的临床安全应用提供理论依据。 Objective To explore the mechanism of Yinchenhao Tang was used to the method of serum metabonomics in the prevention and treatment of(NAFLD) in non-alcoholic fatty liver, and to explore its related metabolic pathway by looking for endogenous potential biomarkers with biological significance in this study. Methods The NAFLD model was established in rats induced by high-fat diet. After the model was successfully established, 3.6 g/kg Yinchenhao Tang was given for 2 weeks. The serum metabonomics of rats in blank group, model group and Yinchenhao Tang group were analyzed by UPLC-Q-TOF-MS technique, and the potential key biomarkers were explored by principal component analysis. The corresponding metabolic pathways were found by combining human metabonomics database(HMDB) and Jingdu gene and genome encyclopedia database(KEGG). Results Yinchenhao Tang could improve the deviation of metabolic track in NAFLD model rats, and find 11 biomarkers which were different from NAFLD model and treated with Yinchenhao Tang, involving 10 related metabolic pathways. By using the analysis of related metabolic pathways, it was found that the four metabolic pathways most widely affected were glycerophospholipid metabolism, linoleic acid metabolism, α-linolenic acid metabolism and nicotinate and nicotinamide metabolism. Conclusion Therefore, it is inferred that Yinchenhao Tang can regulate the serum metabolites of NAFLD model rats, so as to provide theoretical basis for the clinical safety of Yinchenhao Tang in the treatment of related diseases.
