海岛棉GbCHI基因启动子的克隆及瞬时活性表达分析 被引量:4

Cloning and Transient Expression Analysis of the Promoter of Chalcone Isomerase Gene GbCHI from Gossypium barbadense L.

摘要 查尔酮异构酶(chalcone isomerase,CHI)是类黄酮代谢途径中关键的限速酶之一,与植物的花色形成、抗虫、抗病等特性密切相关。本研究利用前期转录组测序及差异基因表达分析,筛选到与海岛棉抗枯萎病相关基因GbCHI,并从海岛棉抗病品种‘06-146’基因组中克隆了该基因的启动子,序列长度为2062 bp。启动子分析软件Plant CARE预测GbCHI基因启动子序列除了含有启动子基本元件TATA-box和CAAT-box外,还包括与光照、激素、真菌诱导响应及参与黄酮类生物合成基因调控的MYB转录因子结合位点等顺式作用元件。通过瞬时转化烟草叶片的表达分析进一步表明:在MeJA、SA和枯萎病诱导下该启动子具有驱动GUS基因表达的特性。本研究结果可为后期开展GbCHI基因参与海岛棉抗枯萎病的调控机制研究奠定良好的理论基础。 Chalcone isomerase(CHI) is one of the key rate-limiting enzymes in the flavonoid metabolism pathway,which is closely related to the flower formation of plants, insect resistance and disease resistance of plants. In this study, based on the previous transcriptome sequencing and differential gene expression analysis, the GbCHI gene related to Fusarium wilt resistance was selected, and the promoter of this gene was cloned from the genome of ’06-146’, a disease-resistant variety of Gossypium barbadense L., with a sequence length of 2 062 bp. Analysis of the GbCHI gene promoter sequence by Plant CARE revealed it contained not only the promoter basic elements TATA-box and CAAT-box, but also includes cis-acting elements such as the response to light, hormones, fungal induction and MYB transcription factor binding sites involved in the regulation of flavonoid biosynthesis genes.The transient expression analysis of tobacco leaves further showed that this promoter has the characteristics of driving the expression of GUS gene under the induction of MeJA, SA and Fusarium wilt. The results of this study can lay a good theoretical foundation for the later research on the regulatory mechanism of GbCHI gene involved in the resistance to Fusarium wilt in Gossypium barbadense L..
