3D biological printing breast cancer model and adriamycin drug sensitivity test

摘要 目的探讨3D生物打印技术构建的乳腺癌模型用于阿霉素药敏试验的可行性。方法通过3D生物打印技术构建MCF-7细胞和MDA-MB-231细胞3D水凝胶乳腺癌模型,分别采用CCK-8法和Live/Dead染色法检测不同浓度阿霉素对2D/3D乳腺癌细胞增殖作用的影响并进行比较。结果2D培养中形成单层片状细胞。而3D打印的乳腺癌模型中MCF-7细胞和MDA-MB-231细胞显示多层结构,细胞多数呈圆形、或梭形和铺展状。MDA-MB-231细胞对阿霉素的耐药性高于MCF-7细胞。3D培养的MCF-7、MDA-MB-231细胞比2D培养对阿霉素的耐药性显著更高。结论3D乳腺癌模型可以在体外更有效、更准确地评估抗癌药物的疗效,可在动物模型或临床试验前为改进和优化药物筛选系统提供有价值的信息。 Objective To explore the feasibility of breast cancer model constructed by 3D bio-printing technology for doxorubicin drug sensitivity test.Methods MCF-7 cells and MDA-MB-231 cells 3D hydrogel breast cancer models were constructed by 3D bio-printing technology.CCK-8 method and Live/Dead staining method were used respectively to detect the effects of different concentrations of adriamycin on the proliferation of 2D/3D breast cancer cells and to make a comparison.Results Monolayer and lamellar cells were formed in 2D culture.MCF-7 cells and MDA-MB-231 cells in the 3D-printed breast cancer model showed multilayer structures,most of which were round or spindle shaped and spread out.MDA-MB-231 cells were more resistant to adriamycin than MCF-7 cells.MCF-7 and MDA-MB-231 cells cultured in 3D were more resistant than those cultured in 2D.Conclusion 3D breast cancer model can more effectively and accurately evaluate the efficacy of anticancer drugs in vitro,which can provide valuable information for improving and optimizing drug screening systems before animal models or clinical trials.
