生物样本中糖胺聚糖定量方法及应用进展

Progress on the Quantitative Methods and Applications of Glycosaminoglycans in Biological Samples

摘要 糖胺聚糖(GAGs)是一类结构复杂、分子量不均一的内源性生物大分子多糖,具有抗凝血、抗肿瘤、抗病毒、调节免疫等多种生物活性,也是多种病理诊断的重要生物标志物,应用前景日益广阔。但由于结构复杂性,GAGs类药物生物样本中的含量测定仍是分析工作的难点,这在一定程度上制约了此类药物的开发应用。文章综述了近年来GAGs类药物生物样本中几种定量方法,包括光谱法、色谱法、毛细管电泳法、放射性标记法,免疫分析法,质谱法等,通过介绍各方法的原理、应用及优缺点,以期对GAGs类药物相关的临床病理诊断、药物监测及药代动力学等研究提供分析工具参考。 Glycosaminoglycan( GAGs) are one kind of endogenous macromolecular polysaccharides with complex structures and heterogeneous molecular weights,they have various biological activities,such as anticoagulation,antineoplastic,antiviral and immunoregulation activities.And they are also important biomarkers formultiple pathological diagnosis.Therefore,application prospect of them are increasingly broad. However,due to the complexity of the structures,there are still difficulties to analyze quantitatively GAGs in the biological samples,which restrict the developments and applications of them to some extent.Several quantitative methods of GAGs in biological samples are reviewed in this paper,including spectroscopy,chromatography,capillary electrophoresis,radiometric labeling,immunoassay,biosensor,and mass spectrometry. We hope to provide analysis tools references of GAGs related clinical pathological diagnosis,medicine monitoring and pharmacokinetic studies by introducing the principles,applications,advantages and disadvantages of each method.
