摘要
文章旨在研究龙生蛭胶囊对高甘油三酯血症模型小鼠的治疗作用,并探索相关作用机制。8周龄C57BL/6小鼠随机分为3组(每组8只小鼠),分别为对照组、低剂量组和高剂量组。对照组喂食高脂食物2周诱导高甘油三酯血症模型;给药组喂食含龙生蛭的高脂食物,低剂量组为每100 g高脂食物中含850 mg龙生蛭,高剂量组为每100 g高脂食物中含2 g龙生蛭。结果显示,给药能够显著降低小鼠血清和肝脏中甘油三酯(triglyceride,TG)的量。机制上,龙生蛭胶囊显著抑制TG合成相关基因二酰甘油酰基转移酶(diacylglycerol o-acyltransferase1,DGAT1)、脂肪酸合酶(fatty acid synthase,FASN),上调TG水解关键基因脂蛋白酯酶(lipoprotein lipase,LPL)的表达。结果证明了龙生蛭胶囊通过抑制TG合成关键基因、促进TG水解相关基因的表达,发挥了治疗高甘油三酯血症的作用。
This paper aims at investigating the therapeutic effect and mechanism of LongShengZhi capsule(LSZ)on hypertriglyceridemia in mice.Eight-week-old C57BL/6 mice were randomly divided into three groups(eight mice in each group):control group,low dose group and high dose group.The mice in control group were fed high-fat diet for two weeks to induce hypertriglyceridemia.The mice in low or high dose group were fed high-fat diet containing 850 mg LSZ per 100 g food or 2 g LSZ per 100 g food.Compared with the control group,LSZ significantly reduced the serum triglyceride(TG)level and liver TG content,and inhibited the TG synthesis related genes diacylglycerol o-acyltransferase1(DGAT1)and fatty acid synthase(FASN),and up-regulated the expression of lipoprotein lipase(LPL)which plays a key role in TG hydrolysis.The results show that LSZ can reduce TG accumulation in both serum and liver by inhibiting gene expression related to TG synthesis and increasing gene expression related to TG hydrolysis.
