摘要
潮间带是水产养殖的重要区域,也是海洋塑料垃圾聚集区,塑料及其表面附着的微生物对养殖生态环境有着重要的影响。本研究利用高通量测序方法,经过基因组DNA提取、PCR产物扩增纯化、构建文库以及测序数据分析,分析了三亚湾潮间带塑料表面附着细菌菌群的结构特征。测序注释结果表明:三亚湾潮间带塑料样本中的细菌分布在28个菌门中,其中平均丰度最高的5个菌门依次为变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)、浮霉菌门(Planctomycetes)、放线菌门(Actinobacteria)、蓝细菌门(Cyanobacteria)。丰度较高的5个属分别为赤杆菌属(Erythrobacter)、鲁杰氏菌属(Ruegeria)、雷辛格氏菌属(Leisingera)、宽球藻属(Pleurocapsa)和梨形菌属(Blastopirellula)。
Intertidal zone is an important area of aquaculture,and as well as a gathering area of marine plastic debris.Plastics and the microorganisms attached to its surface have an important impact on the ecological environment in aquaculture.Different kinds of flora attached to plastic surface in the intertidal zone of Sanya Bay were studied and compared by high throughput sequencing.The structural characteristics of flora attached to plastic surface were studied by genomic DNA extraction,PCR product amplification and purification,constructing the library and analysis of sequencing data.The species annotation of the high throughput sequencing experiment showed 28 phyla in the plastic samples from the intertidal zone of Sanya Bay,among which the five phyla with the highest average abundance are Proteobacteria,Bacteroidetes,Planctomycetes,Actinobacteria and Cyanobacteria,in descending order.The five genera with the highest abundance in the samples are Erythrobacter,Ruegeria,Leisingera,Pleurocapsa and Blastopirellula.
