摘要
目的探讨趋化因子(C-X-C基序)受体4(CXCR4)在乳腺癌骨转移患者乳腺组织中的表达水平及其临床意义。方法收集正常乳腺组织(正常对照组)、乳腺癌无骨转移乳腺组织(乳腺癌无骨转移组)、乳腺癌骨转移乳腺组织(乳腺癌骨转移组)各100例。采用逆转录PCR方法检测乳腺组织的CXCR4 mRNA表达情况,采用免疫组织化学法检测乳腺组织CXCR4的阳性表达情况,比较3组研究对象乳腺组织CXCR4的阳性表达情况和CXCR4 mRNA相对表达量,分析乳腺癌患者乳腺癌组织CXCR4的阳性表达情况与临床病理特征的关系。结果3组研究对象乳腺组织CXCR4的阳性表达情况,差异具有统计学意义(P<0.05),乳腺癌骨转移组患者乳腺组织CXCR4的阳性表达情况最强。乳腺癌患者乳腺癌组织CXCR4的阳性表达情况与糖类抗原153、雌激素受体、孕激素受体、人类表皮生长因子2、肿瘤原发灶大小无关(均P>0.05),而与细胞核相关抗原(Ki-67)、肿瘤分期相关(均P<0.05)。乳腺组织的CXCR4 mRNA的相对表达量由高到低为乳腺癌骨转移组>乳腺癌无骨转移组>正常对照组(均P<0.05)。结论CXCR4在乳腺癌组织中的表达升高,且在乳腺癌骨转移患者乳腺组织中的表达更高,其有可能成为乳腺癌骨转移治疗的新靶点。
Objective To investigate the expression level and clinical significance of chemokine(C-X-C motif)receptor 4(CXCR4)in breast tissue of patients with bone metastasis from breast cancer.Methods Totally 100 cases of normal breast tissue(normal control group),100 cases of breast cancer tissue without bone metastasis(breast cancer without bone metastasis group),and 100 cases of breast cancer tissue with bone metastasis(bone metastasis from breast cancer group)were collected.The expression of CXCR4 mRNA in breast tissues was detected by reverse transcription PCR,the positive expression of CXCR4 in breast tissue was detected by immunohistochemistry,the positive expression of CXCR4 and the relative expression of CXCR4 mRNA in breast tissue were compared among the three groups,and the relationship between the positive expression of breast cancer tissue CXCR4 and clinicopathological characteristics was analyzed in breast cancer patients.Results There was statistically significant difference in positive expression of breast tissue CXCR4 among the three groups(P<0.05),and the maximum positive CXCR4 expression in breast tissue was observed in the bone metastasis from breast cancer group.In breast cancer patients the positive expression of CXCR4 in breast cancer tissue did not correlate with carbohydrate antigen 153,estrogen receptor,progestogen receptor,human epidermal growth factor 2,or size of primary tumor(all P>0.05),but correlated with nucleus-associated antigen(Ki67)and tumor stage(all P<0.05).The relative expression of CXCR4 mRNA in breast tissue decreased in the order of the bone metastasis form breast cancer group,the breast cancer without bone metastasis group and the normal control group(all P<0.05).Conclusion CXCR4 expression is elevated in breast cancer tissue,and the expression is much higher in breast tissue of patients with bone metastasis from breast cancer,which may become a novel target for treating bone metastasis from breast cancer.
