摘要
目的 miR-411-5p对胃癌SGC-7901细胞生物学特性及裸鼠成瘤的影响.方法 荧光定量PCR(RT-qPCR)检测转染效率.5-溴脱氧尿嘧啶核苷(BrdU)染色检测增殖.流式细胞术检测凋亡.干细胞成球实验检测细胞干样特性.蛋白印迹实验检测增殖细胞核抗原(PCNA)、凋亡抑制蛋白(Survivin)、半胱天冬酶-3(Caspase-3)、半胱天冬酶-9(Caspase-9)及P21蛋白的表达.免疫组化检测肿瘤组织中PCNA和Caspase-3的表达.结果 与对照组比较,miR-411-5p过表达组BrdU阳性细胞减少,PCNA和Survivin蛋白表达下调;细胞凋亡率降低,Caspase-3、Caspase-9及P21蛋白表达上调;干细胞数减少.与miR-NC组比较,肿瘤体质量和肿瘤体积减小,肿瘤组织中PCNA和Caspase-3表达下调.均具有显著性差异(P<0.05).结论 miR-411-5p过表达可抑制胃癌SGC-7901细胞增殖、干细胞样特性,促进细胞凋亡,并抑制裸鼠移植瘤生长.
Objective To investigate the effect of Mir-411-5p on the biological characteristics of gastric cancer SGC-7901 cell and tumor formation in nude mice.Methods RT-qPCR was used to detect transfection efficiency.BrdU staining was used to detect proliferation.Flow cytometry was used to detect apoptosis.Stem cell pellet-forming assay was used to detect the characteristics of dry cells.The expression of PCNA,Survivin,Caspase-3,Caspase-9 and P21 proteins were detected by western blot.The expression of PCNA and Caspase-3 in tumor tissues was detected by immunohistochemical assay.Results Compared with the control group,when miR-411-5p overexpressedBrdU positive cells were decreased,and PCNA and Survivin protein expression were down-regulated.The apoptosis rate was decreased,and the expressions of caspase-3,Caspase-9 and P21 were increased.The diameter of stem cells into spheres decreased,and the number of spheres decreased.Compared with miR-NC group,tumor mass and tumor volume decreased.The expression of PCNA and Caspase-3 in tumor tissues was down-regulated.Conclusion The overexpression of miR-411-5p can inhibit the proliferation and stem-like characteristics of gastric cancer SGC-7901 cells,promote cell apoptosis,and inhibit the growth of xenografts in nude mice.
