盐度对凡纳滨对虾血细胞吞噬与免疫相关因子基因表达的影响 被引量:8

摘要 在水温27~29℃下,将平均体长7.01 cm的凡纳滨对虾Litopenaeus vannamei饲养在730 mm×530 mm×450 mm的水族箱中,每箱30只,盐度6、18和30,每个盐度组3个平行,饲养6周,探讨盐度对凡纳滨对虾血细胞吞噬与免疫相关因子基因表达的影响。结果表明:盐度30和18组凡纳滨对虾血细胞吞噬活性显著高于盐度6组对虾(P<0.05),盐度18组血细胞吞噬活性最高。30和18盐度组凡纳滨对虾肝胰腺中Toll样受体(TLR)、免疫缺陷基因(immune deficiency,IMD)、酚氧化酶(PO)、过氧化氢酶(CAT)及溶菌酶(LZM)基因表达水平显著高于盐度6组(P<0.05);18盐度组凡纳滨对虾血细胞中TLR、IMD、PO、SOD及LZM基因表达显著高于6盐度组(P<0.05);盐度30组对虾的TLR基因表达水平显著高于盐度18组(P<0.05),而盐度30组对虾的PO和CAT基因表达水平显著低于盐度18组(P<0.05);其他免疫因子基因的表达水平没有显著性差异。综上,环境中盐度的变化可能通过影响凡纳滨对虾各组织的抗氧化能力、溶菌活性、识别能力和血细胞吞噬活性等,调控凡纳滨对虾细胞和体液免疫。 Pacific white leg shrimp Litopenaeus vannamei with an average body length of 7.01 cm was reared in a 730 mm×530 mm×450 mm aquarium at stocking density of 30 shrimp per aquarium at salinity of 6,18 and 30 at a water temperature of 27~29℃with triplication for 6 weeks to investigate the effects of salinity on blood cell phagocytosis and immune-associated factor gene expression.It was found that there was significantly higher lymphocytes phagocytosis activity in Pacific white leg shrimp exposed to salinity 30 and 18 than that in the salinity 6 group(P<0.05),the maximal lymphocytes phagocytosis activity in salinity 18 group.The expression levels of Toll-like receptor(TLR),IMD,phenol oxidase(PO),catalase(CAT),and lysozyme(LZM)genes were shown to be significantly higher in the hepatopancreas of Pacific white leg shrimp exposed to salinity 30 and 18 than those in the shrimp in 6-salinity group(P<0.05),and expression levels of TLR,IMD,and PO genes significantly lower in the hepatopancreas of Pacific white leg shrimp exposed to salinity 30 than those in the 18-salinity group(P<0.05).The TLR gene expression level was significantly higher in the shrimp in the 30-salinity group than that in the salinity 18 treatment group(P<0.05),while the PO gene expression level of the shrimp in the salinity 30 group was significantly lower than that in the salinity 18 group(P<0.05).In the salinity 30 treatment group,the expression levels of TLR and CAT genes were significantly higher in the gill tissue of Pacific white leg shrimp than those in the salinity 6 group(P<0.05),and the expression levels of CAT and LZM genes were significantly higher in the gill tissue of shrimp in the salinity 18 group than those in the salinity 6 group(P<0.05).The expression level of TLR gene was significantly higher in the gill tis sue of the shrimp in the 30 salinity group than that in the shrimp in the 18 salinity group(P<0.05),while the expression level of the CAT gene was significantly lower in the shrimp in the salinity 30 group than that in the salin
