摘要
壳聚糖是一种从海洋生物中分离得到的甲壳素多糖,具有多种优良特点,被广泛应用于各个领域。随着壳聚糖产品种类的逐渐增多,壳聚糖含量的快速测定标的变得十分重要。采用共振光散射法,选用酸性三苯甲烷类染料测定胶囊中壳聚糖的含量。研究结果表明,壳聚糖在pH为3.20的Clark-Lubs缓冲溶液中可与固绿FCF发生离子缔合反应,生成物可造成共振光散射强度显著增强。在324 nm处共振光强度值ΔI与壳聚糖质量浓度在0.10~1.20 mg/L内呈线性关系,相关系数R为0.9993,检出限为0.015 mg/L。方法用于壳聚糖胶囊的分析,测定值与考马斯亮蓝R250共振光散射法结果一致。
Chitosan is a chitin polysaccharide isolated from a variety of marine organisms.It has a variety of excellent characteristics and is widely used in various fields.Nowadays,products related to chitosan are gradually increasing,so it is very important to determine the content of chitosan in the product.In this experiment,the resonance light scattering method was used to determine the content of chitosan in the capsule with acid triphenylmethane dyes.The results of the study show that chitosan can undergo ion association reaction with Fast Green FCF in Clark-Lubs buffer solution at pH of 3.20,and the resulting product can significantly increase the intensity of resonance light scattering.The resonance light intensity valueΔI at 324 nm has a linear relationship with the concentration of chitosan within 0.10—1.20 mg/L,the correlation coefficient R is 0.9993,and the detection limit is 0.015 mg/L.The method is used for the analysis of chitosan capsules,and the measured value is consistent with the result of Coomassie Brilliant Blue R250 resonance light scattering method.
