摘要
目的基于盒转录因子9(Sox-9)分泌情况,观察柚皮苷刺激骨髓间充质干细胞结合基质载体对兔关节软骨的损伤修复情况。方法对骨髓间充质干细胞进行体外增殖,复合于脱细胞真皮基质载体,植入软骨损伤处,并以柚皮苷汤或去离子水灌胃,设立模型组(Mod组)、骨髓间充质干细胞结合基质载体组(Tec组)、柚皮苷复合骨髓间充质干细胞结合基质载体组(Nar/Tec组),于第12周后对修复组织进行形态学评分、病理学染色及免疫组化染色观察。结果(1)形态学观察上,应用国际软骨修复学会软骨修复的宏观评价量表进行量化评估,Nar/Tec组与其他两组相比,取得了更优的修复效果,且差异具有统计学意义(P<0.05)。(2)病理染色观察上,阿斯新兰染色及番红-O染色,Nar/Tec组在软骨基质及软骨细胞上均可见到明显的修复表现,修复处组织与正常组织相似,Tec组修复效果次之。(3)免疫组化染色观察上,Nar/Tec组在Ⅱ型胶原表达上,优于其他两组;该组在软骨损伤修复处可见明显分泌的Sox-9。结论骨髓间充质干细胞结合基质载体对于兔膝关节软骨损伤有修复作用,加用柚皮苷能提高软骨损伤的修复质量,该效果的产生可能与柚皮苷促进损伤处Sox-9的分泌有关。
Objective Based on SOX-9 secretion,naringin combined with tissue engineering technique was observed to repair the defect of rabbit articular cartilage.Methods Rabbit bone marrow mesenchymal stem cells were proliferated in vitro,compound in acellular dermal matrix carrier,implanted in cartilage defect,and irrigated with naringin soup or deionized water.Model group(Mod group),tissue engineering group(Tec group),naringin binding tissue engineering group(Nar/Tec group)were set up to observe the repair tissue morphologically,pathologically and immunohistochemically after the eighth week.Results(1)In morphological observation,the macroscopic evaluation scale of cartilage repair of the International Cartilage Repair Society(ICRS)was used for quantitative evaluation.The compound naringin group obtained the best repair effect,and the difference was statistically significant(P<0.05).(2)On pathological staining,Asxinlan staining and safranin O staining were similar to those of normal tissues in cartilage matrix and chondrocytes.(3)On immunohistochemical staining,the complex naringin group showed better repair ability in type II collagen,increased Sox-9 secretion was observed.Conclusion Naringin combined with tissue engineering technology can repair cartilage defect of rabbit knee joint naringin’s ability to improve cartilage repair quality may be related to promoting Sox-9 secretion.
