摘要
目的:以芍药苷、阿魏酸及藁本内酯为指标成分,采用体外透皮实验,考察逐瘀止痛远红外灸贴的透皮性能。方法:以3种成分的24 h累积透过量(Q_(24))及渗透倍数为指标对制剂中所用的氮酮的量及接受介质的浓度进行考察。在此基础上测定逐瘀止痛远红外灸贴中芍药苷、阿魏酸及藁本内酯的Q_(24)。并通过测定2、6、8、12、16、24 h的Q_(24)进行各成分的渗透动力学模型拟合。结果:对逐瘀止痛远红外灸贴中3种成分均具有较好作用的促透剂是3%氮酮,最佳的接受介质的浓度为50%生理盐水乙醇溶液。制剂中3种成分的Q_(24)分别为芍药苷26.429μg/cm^(2),阿魏酸13.103μg/cm^(2),藁本内酯5.790μg/cm^(2)。结论:氮酮对该制剂中的3种成分具有明显的促透作用,但并不呈现剂量依赖性。芍药苷、阿魏酸及藁本内酯从逐瘀止痛远红外灸贴中向离体小鼠皮肤渗透符合Higuich方程,具有良好的透皮性能。
Objective:To investigate the transdermal properties of Zhuyu Zhitong Far-Infrared Moxibustion Patch with paeoniflorin,ferulic acid and ligustilide as index components by transdermal test in vitro.Methods:The 24 h cumulative permeation(Q_(24))and permeation multiple of the three components were used as indicators to investigate the amount of azone used in the formulation and the concentration of the receiving medium.On this basis,the Q_(24) of paeoniflorin,ferulic acid and ligustilide in Zhuyu Zhitong Far-Infrared Moxibustion Patch was determined.The permeation kinetics model of each component was fitted by measuring Q_(24) at 2,6,8,12,16 and 24 h.Results:3%azone was the best penetration enhancer for the three components in Zhuyu Zhitong FarInfrared Moxibustion Patch,and the best concentration of the medium was 50%normal saline ethanol solution.The Q_(24) of the three components in the preparation was 26.429μg/cm^(2) of paeoniflorin,13.103μg/cm^(2) of ferulic acid,and 5.790μg/cm^(2) of ligustilide.Conclusion:Azone has a significant effect on promoting penetration of the three components in the preparation,but it does not present a dose-dependent effect.Penetration of paeoniflorin,ferulic acid and ligustilide from Zhuyu Zhitong Far-Infrared Moxibustion Patch into the skin of isolated mice conforms to the Higuich equation and has good transdermal performance.
