龟龄集延缓果蝇衰老的作用研究 被引量:6

摘要 目的采用果蝇生物模型,验证龟龄集延缓衰老的作用。方法本研究采用果蝇寿命实验筛选龟龄集延缓衰老的最佳给药剂量,并确定其最佳给药方式。从果蝇30日龄开始连续给药直至死亡,考察0.5,1,2,4,8 mg/ml共5个不同剂量的龟龄集对果蝇寿命的影响;分别考察连续15 d给药和间隔给药(间隔1 d)15 d对果蝇寿命的影响。通过逆重力爬行实验和抗冷应激实验两个指标评价4 mg/ml龟龄集连续给药15 d抗衰老活性。结果喂养5个不同剂量的龟龄集后,4 mg/ml剂量给药15 d时延长寿命效果较佳。采用4 mg/ml的龟龄集连续给药15 d可显著延长果蝇寿命(P<0.01)。与对照组相比,龟龄集组果蝇逆重力爬行能力差异无统计学意义(P>0.05),而果蝇抗冷应激能力明显提高(P<0.05)。结论龟龄集具有延缓衰老的作用。 Objective To verify the effect of Guilingji on delaying aging in the biological model of Drosophila melanogaster.Methods The different doses of Guilingji(0.5,1,2,4,8 mg/ml)were given to the death in 30-day-old Drosophila melanogaster to evaluate its effect on the lifespan.The effects of administration way,15 d continous adminstration or interval administration(interval of one day),on lifespan were investigated.The anti-aging activity of Guilingji was evaluated by climbing assay and anti-cold stress assay.Results The effect of 4 mg/ml Guilingji for 15 d on mean and median lifespans of Drosophila melanogaster was better.When Drosophila melanogaster were continuously given 4 mg/ml for 15 d,the lifespan was increased significantly(P<0.01).Under the conditions of the above,the athletic ability of Drosophila melanogaster was not improved(P>0.05),while the anti-cold stress ability was improved significantly(P<0.05).Conclusion Guilingji has the effect of delaying aging.
