摘要
目的分析亚洲区西尼罗河病毒(West Nile virus,WNV)的流行情况,探讨其系统发育和分子进化特征。方法从NCBI数据库中分别获取亚洲区流行的WNV全基因组序列和E蛋白序列,进行比对和同源性分析后构建系统进化树,分析来自不同株型病毒的结构蛋白E的核苷酸和氨基酸的突变率,进一步找出氨基酸的替代位点。结果在中国、马来西亚、印度、以色列、巴基斯坦、伊朗和土耳其等地有血清学感染或回顾性检测阳性报道。将获取的WNV全基因组序列进行系统进化树分析,发现WNV以色列株(HM152773、AF481864)在亲缘关系上与谱系1a株最为接近,而以色列株(AY688948)在亲缘关系上与谱系2株较为接近,印度株(KU978770)与谱系1c接近;将获取的WNV E蛋白序列进行系统进化分析,发现WNV中国株(JX442278,JX42280-JX442282)在亲缘关系上与谱系1a株最为接近。氨基酸序列和核酸序列分析表明,不同株型的E蛋白与谱系2相比,表现为碱基和氨基酸的突变率高,与谱系1a相比,印度株(GQ851605、JX041632、GQ851604和KU978770)的第154和第156位氨基酸分别由天冬酰胺突变为天冬氨酸和丝氨酸(N154D,N154S),丝氨酸突变为脯氨酸和丙氨酸(S156P,S156A),而中国株(AY490240)、巴基斯坦株(JX070655)和以色列株(HM051416)的第156位氨基酸分别由丝氨酸突变为脯氨酸、苯丙氨酸和脯氨酸(S156P、S156F、S156P)。结论通过谱系分析、系统进化树构建、包膜蛋白E氨基酸序列及编码核苷酸序列的比较分析发现,有些位点的改变可能与病毒的毒力、致病性、神经侵袭力、流行等有关,为研究亚洲区不同国家不同株型间WNV的生物学和流行致病性差异提供了参考。
Objective To analyze the epidemic of West Nile virus(WNV)in Asia and investigate its phylogenetic and molecular characters.Methods The whole genome sequence and E protein sequence of WNV were retrieved from NCBI database for comparison and homology analysis,based on which a phylogenetic tree was constructed to analyze the mutation rates of viral structural protein E in nucleotides and amino acids from different types of strains and to find out the substitution sites of amino acids.Results There were the positive serological infection or retrospective test reports in China,Malaysia,India,Israel,Pakistan,Iran and Turkey.The phylogenetic tree analysis of WNV whole genome sequence revealed that two WNV Israel strains(HM152773 and AF481864)were the closest to the lineage 1 a strain in the genetic relationship,while one Israel strain(AY688948)was closer to the lineage 2,and the Indian strain(KU978770)was closer to the lineage 1 c.The phylogenetic tree analysis of WNV E protein sequence revealed that two WNV China strains(JX442278 and JX42280-JX442282)were the closest to lineage 1 a.Compared with lineage 2,the mutation rates of nucleotides and amino acids of E protein of different subtypes were high.Compared with lineage 1 a,the amino acids at sites 154 and 156 of Indian strain(GQ851605,JX041632,GQ851604 and KU978770)changed from asparagine to aspartic acid and serine(N154 D,N154 S),and from serine to proline and alanine(S156 P,S156 A),respectively.However,the amino acids at site 156 of Chinese strain(AY490240),Pakistan strain(JX070655)and Israel strain(HM051416)changed from serine to proline,phenylalanine and proline(S156 P,S156 F,S156 P),respectively.Conclusion By pedigree analysis,phylogenetic analysis and comparative analysis of nucleotides and amino acids sequence of envelope protein E,some mutations in the E protein may be related to virulence,pathogenicity,neuroinvasive and epidemic of the virus.This finding provides a reference for the study on biological and epidemiological differences of WNV in Asia.
