共表达网络鉴定食管鳞癌与腺癌关键差异表达分子 被引量:1

Identification of key differential genes between esophageal squamous cell carcinomaand esophageal adenocarcinoma using gene co-expression network

摘要 目的:探究食管鳞癌与食管腺癌关键差异表达分子,为临床个体化治疗和靶向治疗提供依据.方法:应用食管鳞癌和食管腺癌的mRNA转录组数据确定其差异表达基因;根据无标度网络理论及差异表达基因构建加权基因共表达网络;通过相关性分析确定与食管腺、鳞癌分组特征相关性最强的网络模块及模块内节点基因.结果:通过共表达网络构建出19个基因模块,其中Blue和Turquoise模块与食管腺癌、鳞癌相关性最强,相关系数分别为-0.95和0.89;EPS8L3和SOX15分别为Blue和Turquoise模块的代表性节点基因;在TCGA食管癌数据集、GSE26886数据集、癌细胞数据库中,EPS8L3和SOX15基因是食管腺癌和食管鳞癌的关键差异基因,能够鉴别诊断食管鳞癌和食管腺癌.结论:EPS8L3和SOX15可作为食管腺癌和鳞癌关键差异表达分子,为食管癌精准治疗提供理论基础. Objective:Explore the key differentially expressed molecules between esophageal squamous cell carcinoma(ESCC)and esophageal adenocarcinoma(EAC),and provide a basis for clinical individualized treatment and targeted therapy.Methods:The mRNA transcriptome data of ESCC and EAC were analyzed for identification of differentially expressed genes(DEGs).Based on the characteristics of scale-free networks,the DEGs were used to construct the network through weighted gene co-expression network analysis(WGCNA).Afterwards,the modules with the strongest correlation with the characteristics of ESCC/EAC were determined through correlation analysis,and the hub genes of the module were determined by the degree of module membership and gene significance.Results:A total of 19 gene modules were identified through WGCNA.Among them,Blue and Turquoise modules had the strongest correlations with ESCC/EAC with correlation coefficients of-0.95 and 0.89,respectively.EPS8L3 and SOX15 were representative genes of Blue and Turquoise modules,respectively.External validation in data sets of TCGA esophageal cancer,GSE26886,and Cancer Cell Line Encyclopedia showed that these two genes were the key differential genes for ESCC and EAC.Conclusion:EPS8L3 and SOX15 may be used as the key differentially expressed molecules in ESCC and EAC,providing a theoretical basis for precise treatment of esophageal cancer.
