摘要
目的探讨小鼠皮肤/肌肉切口牵拉模型的建立方法及右美托咪定对小鼠疼痛行为学的影响。方法取22只健康雄性C57BL/6小鼠,随机分为假手术组(皮肤/肌肉只切开不牵拉)和实验组(皮肤/肌肉切口牵拉),每组各11只。分别于术前1 d和术后1、3、7、10、14和21 d,通过疼痛行为学检测小鼠机械缩足反射阈值和热缩足潜伏期以评价是否造模成功。另选取48只健康雄性C57BL/6小鼠,随机分为假手术组和实验组,造模方法与上述相同,于术前1 d时行机械痛阈值测试,随后实验组均于腹腔注射0.9%氯化钠注射液和不同剂量的右美托咪定(0.50、2、10μg/kg),观察药物对模型小鼠触诱发痛的影响。结果与假手术组比较,实验组术后1 d开始机械痛阈值显著降低,且持续至术后10 d,随后升高直至术后21 d时恢复术前水平(P<0.05);两组手术前后不同时间点热缩足潜伏期比较差异无统计学意义(P>0.05)。假手术组腹腔注射不同剂量右美托咪定前后机械痛阈值比较差异无统计学意义(P>0.05)。实验组给予右美托咪定(0.50、2μg/kg)30 min后机械痛阈值与给药前比较差异无统计学意义(P>0.05);实验组给予右美托咪定(10μg/kg)30 min后机械痛阈值明显高于给药前(P<0.05)。结论小鼠皮肤/肌肉切口牵拉构建持续性术后疼痛的模型简单方便、安全有效、稳定可行,且通过右美托咪定干预可有效减轻模型小鼠触诱发痛。
Objective To analyze the establishment of skin/muscle incision and retraction model in mice and the effect of dexmedetomidine on pain behavior.Methods 22 healthy male C57BL/6 mice were randomly divided into sham operated group(skin/muscle incision without pulling)and experimental group(skin/muscle incision and retraction).On the 1st day before operation and the 1st,3rd,7th,10th,14th and 21st day after operation,the threshold of mechanical foot contraction reflex and latent period of thermal foot contraction were detected by pain behavior to evaluate whether the model was successful.In addition,healthy male C57BL/6 mice were randomly divided into sham operation group and experimental group.The method of modeling was the same as the above.The mechanical pain threshold was tested one day before operation.Then,the experimental group was injected with 0.9%sodium chloride injection and different doses of right metomidine(0.50μg/kg,2μg/kg,10μg/kg)intraperitoneally to observe the effect of drugs on touch induced pain in model mice.Results Compared with the sham operation group,the mechanical pain threshold of the experimental group decreased significantly at the first day after operation,and continued to 10 days after operation,and then increased until the preoperative level was restored at 21 days after operation(P<0.05).There was no statistical significance between the two groups at different time points before and after operation(P>0.05).There was no significant difference in the mechanical pain threshold between the sham operation group and the sham operation group(P>0.05).There was no significant difference in the mechanical pain threshold between the experimental group and the pre-treatment group(P>0.05)after 30 minutes of administration of dexmedetomidine(0.50μg/kg,2μg/kg);the mechanical pain threshold of the experimental group after 30 minutes of administration of dexmedetomidine(10μg/kg)was significantly higher than that before administration(P<0.05).Conclusion It is simple,convenient,safe,effective,feasib
