正交试验设计优化石墨炉原子吸收法测定孕妇尿锰的方法探讨

摘要 目的选择石墨炉原子吸收光谱法测定尿锰的最佳试验条件,建立尿中锰的测定方法。方法应用L_(16)(4^(3))正交表安排石墨炉原子吸收光谱法测定尿锰时灰化温度、原子化温度及样品稀释液中硝酸的浓度并进行试验,优化各项仪器操作参数,考察灰化温度、原子化温度及各项分析性能指标,包括准确度、精密度、检出限,建立测定尿锰的方法。结果石墨炉原子吸收光谱法测定尿中锰的最佳条件:灰化温度和原子化温度分别为1400℃和1900℃,分别测定加标样品的相对标准偏差RSD值(n=8)在0.65%~4.78%,样品加标回收率在100.2%~113.7%,检出限为0.06μg/L。结论利用正交试验设计优化试验条件在减少检测量同时,可以提高质量,获得的分析方法准确、灵敏,符合尿中锰的测定要求。 Objective To explore the best laboratory condition for the determination Manganese in urine by Graphite Furnace Atomic Absorption Spectrum(GFAAS).Methods The three major factors including ashing temperature,atomization temperature and different concentration of sample dissolved HNO3 were arranged and tested according to the L_(16)(^(43))orthogonal table.The operation conditions of the apparatus were optimized and the performances were estimated including accuracy precision and detection limit.Results The best ashing temperature and atomization temperature were 1,400℃and 1,900℃respectively;the RSD were 0.65%to 4.78%,the recoveries were 100.2%to 113.7%and the detection limit was 0.06μg/L.Conclusion Efficient experiment and reasonable results could be obtained with the orthogonal experiment design.Using the orthogonal test design to optimize the experimental conditions can reduce the amount of detection,improve the quality,obtain accurate and sensitive analytical methods,meet the requirements of the determination of manganese in urine.
