Role of NLRP3 inflammasome in airway remodeling in rats with chronic obstructive pulmonary disease

摘要 目的探讨Nod样受体蛋白-3(NLRP3)炎性小体在慢性阻塞性肺疾病(慢阻肺)大鼠气道重塑中的作用。方法用随机数字表法将24只Wistar大鼠均分为慢阻肺组及空白组(各12只),模型制造成功后测定两组大鼠外周血淋巴细胞NLRP3 mRNA、肺泡灌洗液炎性因子、气管平滑肌厚度水平。结果慢阻肺组外周血淋巴细胞NLRP3 mRNA水平高于空白组(P<0.05)。慢阻肺组气管平滑肌厚度为(14.25±0.63)μm^(2)/μm,空白组为(10.44±0.61)μm^(2)/μm,慢阻肺组高于空白组(P<0.05)。与空白组相比,慢阻肺组肺泡灌洗液的IL-18[(62.98±1.69)比(36.24±3.45)ng/L]、慢阻肺组肺泡灌洗液的IL-1β[(54.51±1.91)比(38.06±0.90)ng/L]水平升高(均P<0.05)。气管平滑肌厚度与外周血淋巴细胞NLRP3 mRNA相对表达量的r值为0.799;与肺泡灌洗液IL-18的r值为0.938;与肺泡灌洗液IL-1β水平的r值为0.970(均P<0.05)。结论慢阻肺大鼠外周血淋巴细胞NLRP3炎性小体表达水平升高,在其气道重塑中发挥关键作用,为治疗慢阻肺提供了新的靶点。 Objective To investigate the role of Nod like receptor protein-3(NLRP3)inflammasome in airway remodeling in rats with chronic obstructive pulmonary disease(COPD).Methods Using the random number table method,24Wistar rats were divided in⁃to COPD group and control group(12 rats each).The levels of NLRP3 mRNA in peripheral blood lymphocyte,inflammatory cytokines in bronchoalveolar lavage fluid and airway smooth muscle thickness were measured after the models were successfully established.Re⁃sults The NLRP3 mRNA level of peripheral blood lymphocytes in COPD group was higher than that in control group(P<0.05).The thickness of airway smooth muscle in COPD group was(14.25±0.63)μm^(2)/μm,which was higher than that in control group[(10.44±0.61)μm^(2)/μm](P<0.05).Compared with the control group,the levels of IL-18 in BALF of COPD Group[(62.98±1.69)vs.(36.24±3.45)ng/L]and IL-1βin BALF of COPD Group[(54.51±1.91)vs.(38.06±0.90)ng/L]were increased(all P<0.05).The R value be⁃tween the thickness of tracheal smooth muscle and the relative expression of NLRP3 mRNA in peripheral blood lymphocytes was 0.799;the R value between the thickness of tracheal smooth muscle and IL-18 in bronchoalveolar lavage fluid was 0.938;the R value between the thickness of tracheal smooth muscle and IL-1βin bronchoalveolar lavage fluid was 0.970(all P<0.05).Conclusion The increased expression of NLRP3 inflammasome in peripheral blood lymphocytes of COPD rats plays a key role in airway remodeling,which provides a new target for the treatment of COPD.
