摘要
数字PCR(digital PCR,dPCR)技术是近几年迅速发展起来的第三代PCR技术,通过把反应体系均分到大量反应室中独立扩增单分子核酸,采集产生的荧光信号后根据泊松分布和阳性比例计算得到检测结果。dPCR技术能实现对样本中低水平目标核酸的绝对定量,具有高灵敏度、高准确度、重现性好、无需标准曲线等优点。本文综述了dPCR的基本原理、主要优势、技术平台发展及其在动物病原体检测、生长繁殖相关基因检测、畜禽饲料成分检测和畜禽产品质量分析等方面的应用。
Digital PCR(dPCR)has been the third generation of PCR technology and developed rapidly in recent years.The mixed PCR reaction system is divided into a large number of partitions where a single molecule is amplified independently.The fluorescence signals are then collected,and the results are calculated according to the Poisson distribution and positive ratio.This technique can realize absolute quantification of target nucleic acid in extremely low concentration,and possesses various advantages such as high sensitivity,high accuracy,good reproducibility,and no need of standard curve.In this paper,the basic principle,main advantages,developed platforms of dPCR and its applications in the detection of animal pathogens,genes related to growth and reproduction,feed ingredients,as well as quality analysis of animal products are reviewed.
